Pengaruh Ekstrak Daun Nicotiana Tabacum Terhadap Sensitivitas Insulin Pada Tikus Model Diabetes Mellitus Tipe II

Abstract

Diabetes mellitus (DM) tipe 2 merupakan penyakit metabolic yang ditandai dengan tingginya gula darah seseorang yang sudah berlangsung kronis, dan biasanya disebabkan oleh gaya hidup orang tersebut. Mekanisme tingginya gula darah pada diabetes melitus tipe 2 yaitu disebabkan berkurangnya produksi insulin di pankreas, berkurangnya sensitivitas insulin, atau kombinasi keduanya. Inflamasi diketahui memiliki kaitan erat dengan DM tipe 2, dimana dengan adanya inflamasi, produksi insulin dan juga sensitivitasnya di jaringan akan menurun. Nikotin diduga memiliki efek anti-inflamasi, dan Indonesia memiliki sumber daya nikotin yang berlimpah (diambil dari daun Nicotiana tabacum). Penelitian mengenai pengaruh ekstrak daun Nicotiana tabacum terhadap sensitivitas insulin belum pernah dilaksanakan, karena itu peneliti bertujuan untuk mengetahui tentang pengaruh pemberian ekstrak daun Nicotiana tabacum terhadap sensitivitas insulin berdasarkan penghitungan HOMA-%S pada model tikus (Rattus norvegicus) galur Wistar yang diinduksi diabetes mellitus tipe 2. Studi eksperimental menggunakan randomized post test only controlled group design. Sampel dibagi menjadi 5 kelompok, kelompok “kontrol negatif”, “kontrol positif”, “perlakuan 1”, “perlakuan 2”, dan “perlakuan 3”. Hasil penelitian menunjukkan bahwa pemberian ekstrak daun Nicotiana tabacum tidak memiliki pengaruh yang bermakna terhadap sensitivitas insulin yang dinilai menggunakan metode HOMA-%S (ANOVA, p=0,362).

English Abstract

Type 2 diabetes mellitus (DM) is a metabolic disease characterized by chronic elevation of blood sugar in a person, and usually caused by the person lifestyle. The mechanism of blood sugar elevation in type 2 diabetes mellitus caused by reduction of the insulin production, decreased in insulin sensitivity, or combination of both. Inflammation is known to have a close relationship with type 2 diabetes, which in the presence of inflammation, insulin production and also its sensitivity in the tissues will decrease. Nicotine is thought to have an anti-inflammatory effect, and Indonesia has an abundant nicotine resource (taken from the leaves of Nicotiana tabacum). Research on the effect of Nicotiana tabacum leaf extract on insulin sensitivity has never been implemented, therefore the researcher aims to know about the effect of Nicotiana tabacum leaf extract on insulin sensitivity based on HOMA-% S calculation on mouse model (Rattus norvegicus) Wistar strain induced type 2 diabetes mellitus. Experimental study using randomized post test only controlled group design. Samples were divided into 5 groups, "negative control", "positive control", "treatment 1", "treatment 2", and "treatment 3" groups. The results showed that Nicotiana tabacum leaf extract had no significant effect on insulin sensitivity assessed using HOMA-% S (ANOVA, p = 0,362).
