Pengaruh Ekstrak Kulit Tomat (Solanum Lycopersicum) Terhadap Kadar Glukosa Darah Puasa Pada Tikus Model Diabetes Melitus Tipe 2

Abstract

Diabetes melitus (DM) adalah sindroma metabolik yang ditandai dengan hiperglikemia. DM tipe 2 disebabkan oleh kelainan resistensi insulin sehingga sel dan jaringan tidak dapat memanfaatkan glukosa akibatnya terjadi hiperglikemia. Keadaan hiperglikemia menyebabkan peningkatan radikal superoksida yang mengakibatkan gangguan pada sel β pankreas. Tomat (Solanum lycopersicum) pada bagian kulit memiliki kandungan antioksidan yang lebih tinggi dibandingkan dengan daging buah maupun tomat utuh seperti likopen, β-karoten, vitamin A, vitamin C, vitamin E, dan kuersetin. Kandungan antioksidan ini dapat menghambat radikal bebas sehingga berpotensi dalam menurunkan kadar glukosa darah puasa. Penelitian ini bertujuan untuk membuktikan pengaruh ekstrak kulit tomat dalam menurunkan kadar glukosa darah puasa dengan menggunakan tikus putih Rattus norvegicus model DM tipe 2. Sampel sebanyak 20 dibagi menjadi lima kelompok yaitu kontrol negatif, kontrol positif, 3 kelompok perlakuan yang diberi ekstrak kulit tomat dosis 50mg/kgBB, 100 mg/kgBB, dan 150 mg/kgBB selama 4 minggu. Kadar glukosa darah diukur sebelum dan sesudah perlakuan. Data kadar glukosa darah puasa diuji dengan Paired T test dan Wilcoxon Signed test dengan signifikansi 0,05. Hasil penelitian menunjukkan pada pemberian esktrak kulit tomat dosis 50 mg/kgBB terjadi penurunan kadar glukosa darah puasa (GDP) yang signifikan, sedangkan pada ekstrak kulit tomat dosis 100mg/kgBB dan 150 mg/kgBB terjadi penurunan kadar GDP namun tidak signifikan. Kesimpulan dari penelitian ini adalah pemberian ekstrak kulit tomat dengan dosis 50 mg/kgBB dapat menurunkan kadar glukosa darah puasa pada tikus model DM tipe 2.

English Abstract

Diabetes Mellitus (DM) is a metabolic syndrome identified by hyperglycemia. DM Type 2 is caused by abnormalities of insulin resistance therefore the cells and membranes fail to utilize glucose and end up with hyperglycemia as the result. The state of hyperglycemia induces an increasing superoxide radicals that caused disruptions in pancreatic β cells. Tomato (Solanum lycopersicum) on the peel has a higher level of antioxidants contents than on the flesh of the fruit or in a whole intact tomato such as lycopene, β-carotene, vitamin A, vitamin C, vitamin E, and quercetin. These antioxidants contents can hold up any free radicals so they were potential in lowering fasting blood glucose levels. This research aims to prove the effects of tomato peel extract in lowering fasting blood glucose levels by using white rats Rattus norvegicus DM type 2 model. Total sample of 20 were divided into five groups which were negative control, positive control, 3 treatment groups that were given a dose of tomato peel extract 50mg/kgBW, 100 mg/kgBW, and 150 mg/kgBW for 4 weeks. Blood glucose levels were measured before and after treatment. Data in fastingblood glucose levels tested with Paired T test and Wilcoxon Signed test with 0.05 significations. The results showed that dose of 50 mg/kgBW tomato peel extract decreased the levels of fasting blood glucose (FBG) significantly, while the dose of 100mg/kgBW and 150 mg/kgBW tomato peel extract decreased levels of FBG less significant. The conclusion of this research is that the tomato peel extract at a dose of 50 mg/kgBW tomato peel extract can lower fasting blood glucose levels in a rat model of DM type 2.
