Abstract

Aedes aegypti sangat penting pada daerah tropis dan subtropis karena merupakan vektor dari dengue yang merupakan masalah kesehatan besar di Indonesia dengan prevalensi dan angka kematian yang tinggi. Penggunaan insektisida sintetik menimbulkan masalah polusi dan resistensi, sehingga dibutuhkan insektisida alami. Cengkeh (Syzygium aromaticum) mengandung saponin dan flavonoid yang dapat menghambat perkembangan telur nyamuk. Tujuan penelitian ini adalah untuk mengetahui ovicidal activity esktrak etanol daun cengkeh terhadap telur Aedes aegypti dan membuktikan adanya kerusakan micropyle. Penelitian ini menggunakan metode eksperimental dengan rancangan true experimental-post test only control group design. Sampel yang digunakan adalah telur Aedes aegypti dengan morfologi yang baik sebanyak 500 butir dibagi menjadi 5 kelompok perlakuan dengan 4 pengulangan. Perlakuan terdiri atas abate sebagai kontrol positif, air sumur sebagai kontrol negatif, dan 3 perlakuan ekstrak daun cengkeh dengan konsentrasi masing-masing 0,5%, 1%, dan 2%. Jumlah telur yang tidak menetas diamati setiap 12 jam selama 48 jam. Selanjutnya dilakukan pengamatan kerusakan telur menggunakan mikroskop. Hasil uji Kruskal-Wallis menunjukkan p sebesar 0,001. Ovicidal activity pada jam ke-48 terbesar adalah 100% yang didapatkan pada konsentrasi ekstrak 2%, diikuti oleh konsentrasi 1% dengan ovicidal activity sebesar 65%, dan konsentrasi 0,5% dengan ovicidal activity sebesar 50%. Kesimpulan penelitian ini adalah ekstrak daun cengkeh memiliki ovicidal activity terhadap telur Aedes aegypti yang berbanding lurus dengan konsentrasi ekstrak dan didapatkan kerusakan micropyle telur Aedes aegypti pada konsentrasi esktrak 2%.

English Abstract

Aedes aegypti is very important in tropical dan subtropical areas because it is the vector of dengue which is a major health problem in Indonesia with high prevalence and death rates. The use of synthetic insecticides have caused problems like polution and resistance, so natural insecticides are in need. Cloves (Syzygium aromaticum) contains falvonoids and saponins which can inhibit the development of mosquito eggs. The purpose of this study was to determine the ovicidal activity of clove leaf ethanol extract against Aedes aegpyti egg and to prove the micropyle damage. This research used experimental method with true experimental design-post test only control group design. The samples were Aedes aegypti eggs with good morphology, 500 were divided into 5 groups with 4 repetitions. The treatment consisted of abate as a positive control, well water as a negative control, and 3 treatment of clove leaf extract with a concentration of 0.5%, 1% and 2% respectively. The number of eggs that did not hatch was observed every 12 hours for 48 hours. After that, the damage rates of eggs were observed using a microscope. Kruskal-Wallis test result showed p value of 0,001. The highest ovicidal activity at 48th hour is 100% which obtained at the extract concentration of 2%, followed by concentration of 1% with ovicidal activity of 65%, and concentration of 0.5% with ovicidal activity of 50%. It is concluded that clove leaf extract has an ovicidal activity on the eggs of Aedes aegypti that proportional to the concentration of extract and micropyle damage is obtained on the egg of Aedes aegypti on treatment with 2% clove leaf extract.
