Abstract
Diabetes Melitus (DM) merupakan penyakit metabolik yang ditandai oleh hiperglikemia. DM tipe 2 disebabkan resistensi insulin. Dalam keadaan hiperglikemia, terjadi proses peningkatan Reactive Oxygen Species (ROS). Tomat (Solanum lycopersicum) memiliki banyak kandungan antioksidan. Superoksida Dismutase (SOD) merupakan antioksidan enzimatik yang mengreduksi ROS dengan mengkatalisa anion superoksida menjadi hidrogen peroksida. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak kulit tomat secara oral terhadap aktivitas SOD tikus model DM tipe 2 dengan studi eksperimental menggunakan rancangan Randomized Post Test Controlled Group Design pada tikus putih Rattus norvegicus galur Wistar jantan. Sampel sebanyak 20 dibagi menjadi lima kelompok yaitu kontrol negatif, kontrol DM tipe 2, kelompok perlakuan DM tipe 2 yang diberi ekstrak kulit tomat selama 4 minggu yang kemudian dilakukan pemeriksaan aktivitas SOD. Hasil penelitian menunjukkan bahwa aktivitas SOD pada kelompok tikus DM tipe 2 lebih tinggi daripada kelompok tikus normal (39,59 U/ml vs 34,2 U/ml). Aktivitas SOD masing – masing kelompok berbeda bermakna dengan p < 0,05. Penurunan aktivitas SOD paling besar didapatkan pada pemberian ekstrak kulit tomat dosis 150 mg/kgBB.
English Abstract
Diabetes Mellitus (DM) is a metabolic disease, characterized with hyperglycemia. Type 2 DM is caused by insulin resistance, which is caused by obesity, sedentary lifestyle, and age. In hyperglycemia state, Reactive Oxygen Species (ROS) level is increased due to the excitation of mitochondrial membrane. Tomato (Solanum lycopersicum) contains many antioxidants. Superoxide Dismutase (SOD), an enzymatic antioxidant in cells, reduces ROS by catalyzing superoxide anion to hydrogen peroxide. This research is intended to understand the effect of tomato skin extract towards SOD activity with true experimental study by using randomized post-test controlled group design in rats type 2 DM models. Total samples are 20, divided by 5 groups: negative control, DM type 2 models and DM type 2 models given therapy for four weeks, afterwards activity of SOD is tested. The result of the experiment shows that the activity of SOD of group of rats DM type 2 higher than the group of normal rats (39,59 U/mL vs 34,2 U/mL). The highest decreasing effect of SOD activity is found in dose 150 mg/kgBW group.
