Pengaruh Pemberian Darapladib Terhadap Jumlah Vasa Vasorum Pada Aorta Tikus Sprague-Dawley Model Aterosklerosis Dengan Dislipidemia

Abstract

Aterosklerosis adalah penyakit inflamasi kronis pada pembuluh darah yang dapat menyebabkan berbagai manifestasi penyakit kardiovaskuler. Salah satu faktor resiko utama aterosklerosis yaitu dislipidemia. Peningkatan profil lipid akan memacu terjadinya disfungsi endotel, sehingga terjadi inflamasi dengan rekuitment monosit, makrofag, limfosit T, dan sel mast yang akan mensekresikan enzim Lp-PLA2 dan berikatan dengan LDL teroksidasi akibat pengaruh ROS (Reactive Oxygen Species). Kemudian makrofag berikatan dengan Ox-LDL menjadi foam cell, migrasi otot polos. Reseptor Lp-PLA2 berperan menghidrolisis Oxidized Phosphatidylcholine (Ox-PC) menghasilkan senyawa bioaktif yaitu Lysophosphatidylcholine (LysoPC) dan Oxidized Nonesterified Fatty Acids (Ox-NEFA). Hal ini berpengaruh terjadinya hipoksia jaringan yang menyebabkan peningkatan angiogenesis vasa vasorum, lalu berubah menjadi immatur, mudah ruptur sehingga berpengaruh terhadap progresifitas plak aterosklerosis. Studi experimental menggunakan Randomized Only Post-Test with Control Group Design yang terbagi menjadi 2 serial waktu. Masing-masing kelompok terdiri dari normal, dislipidemia, dan dislipidemia yang diberikan Darapladib (n=5) sehingga total 30 tikus. Variabel yang diukur yaitu jumlah vasa vasorum setiap kelompok. Tujuan penelitian ini yaitu ingin mengetahui pengaruh pemberian Darapladib dosis 20mg/KgBB tikus dalam menurunan jumlah vasa vasorum kelompok perlakuan 8 dan 16 minggu yang diberikan high fat diet (HFD). Hasil penelitian menunjukkan hasil perbedaan signifikan antara kelompok (p< 0,05). Kesimpulan dari penelitian ini adalah pemberian Darapladib memiliki pengaruh terhadap penurunan jumlah vasa vasorum tikus yang diberi HFD. Peneliti menyarankan agar mengetahui lebih lanjut mengenai variasi dosis efektif dan uji toksisitas pada hewan coba maupun manusia.

English Abstract

Atherosclerosis is a chronic inflammatory disease of the blood vessels that responsible to cause variations manifestation of cardiovascular disease. One of the main risks factors of atherosclerosis is dyslipidemia. The increasing of lipid profile absolutely spurs the occurence of endothelial dysfunction, so it happens with inflammatory recruitment monocytes, macrophages, T lymphocytes and mast cells will secrete the enzyme Lp-PLA2 and Oxidized LDL bound to result ROS (Reactive Oxygen Species). The macrophage will bind with Ox-LDL became foam cells, smooth muscle migration. Lp-PLA2 has a role on Oxidized phosphatidylcholine (Ox-PC) hydrolysis, and bioactive such as Lysophoshatidlycholine (LysoPC) and Oxidized Nonesterified fatty acids (Ox-NEFA).This effect on the level of tissue hypoxia, which causes angiogenesis vasa vasorum mount also became immatured, and easily rupture, which can improve the progressivity plaque of atherosclerosis. Experimental study using designed and it was run in Randomized Only Post-Test Control Group Design was divided into 2 series time 8 and 16 weeks. Each group consists of normal, dyslipidemia, and dyslipidemia was given Darapladib (n=5) with a total of 30 rats. The variable which be measured is the number of vasa vasorum in each group. The purpose of this research was to explore the effect of giving Darapladib with a dose of 20mg/kg in the body weight of rat to reducing the number of vasa vasorum that treatment group 8 and 16 weeks were given a high fat diet (HFD). The result showed significant difference among all groups (p< 0.05). The conclusion is Darapladib has a big influence toward the decreasing of vasa vasorum rat numbers. Further study needed to know the variations in effect dose and toxicity tests in animals and humans.
