Abstract

Kondisi hiperglikemia yang berkepanjangan dapat menimbulkan berbagai macam komplikasi seperti penyakit jantung, stroke, diabetic retinophaty, gagal ginjal, dan gangguan aliran darah pada anggota gerak yang dapat memicu terjadinya amputasi. Dan saat ini, jumlah kasus obesity-linked type-2 diabetes terus meningkat, sehingga banyak dilakukan penelitian tentang manfaat tanaman herbal dalam mengatasi hiperglikemia dan komplikasinya, salah satunya adalah tanaman dengan kandungan antosianin. Penelitian ini bertujuan membuktikan bahwa antosianin dari ubi ungu kultivar Gunung Kawi dapat menurunkan kadar glukosa darah yang meningkat akibat kejadian obesitas. Studi eksperimental laboratorium dilakukan terhadap hewan coba zebrafish. Sampel dipilih dengan cara random sampling untuk dibagi dalam lima kelompok, yaitu kelompok DIO (Diet Induced Obesity) A dengan paparan antosianin 80ppm (n=5), kelompok DIO (Diet Induced Obesity) B dengan paparan antosianin 120ppm (n=5), kelompok DIO (Diet Induced Obesity) C dengan paparan antosianin 160ppm (n=5), kelompok kontrol positif/DIO (n=5), dan kelompok kontrol negatif/non-DIO (n=5). Variabel yang diukur adalah kadar glukosa darah dengan menggunakan alat handheld glucose meter. Hasil analisis data dengan One-way ANOVA menunjukkan bahwa tidak terdapat perbedaan yang signifikan terhadap semua kelompok perlakuan, dengan p=0.119. Kesimpulan dari penelitian ini adalah pemaparan antosianin ubi ungu (Ipomoea batatas L) pada zebrafish (Danio rerio) dengan diet-induced obesity (DIO) tidak dapat menurunkan kadar glukosa darah, namun menunjukkan kecenderungan penurunan kadar glukosa darah.

English Abstract

Prolonged hyperglycemia condition can lead to various complications such as heart disease, stroke, diabetic retinophaty, kidney failure, and impaired blood flow to the limbs that can lead to amputation. As the number of cases of obesity-linked type-2 diabetes continues to rise, a lot of researches about the benefits of herbal plants in treating hyperglycemia and its complications done, one of which is a plant with anthocyanin content. This study aims to prove that anthocyanins from purple sweet potato cultivars Gunung Kawi can lower blood glucose levels rise due to obesity. Laboratory experimental studies conducted on zebrafish. Samples were selected by random sampling to be divided into five groups, namely the DIO (Diet Induced Obesity) A with exposure to 80 ppm anthocyanin (n = 5), group DIO (Diet Induced Obesity) B with exposure to 120 ppm anthocyanin (n = 5), group DIO (Diet Induced Obesity) C with exposure to 160 ppm anthocyanin (n = 5), the positive control/ DIO group (n = 5), and a negative control / non-DIO group (n = 5). The variables measured were blood glucose levels using a handheld glucose meter. The results of the data analysis by One-way ANOVA showed that there was no significant difference to all treatment groups, with p = 0.119. The conclusion of this study is exposure of purple sweet potato (Ipomoea batatas L) anthocyanin in zebrafish (Danio rerio) with diet-induced obesity (DIO) can not lower blood glucose levels, but showed a tendency of decrease in blood glucose levels.
