Abstract
Lupus eritematosus sistemik merupakan suatu penyakit autoimun yang membahayakan. Berbagai upaya terapi telah dikembangkan untuk membantu mencegah progresivitas penyakit ini. Salah satu terapi yang kini dikembangkan adalah terapi dengan menggunakan vaksin kinoid. Vaksin kinoid merupakan vaksin yang bertujuan untuk menginduksi antibodi yang dapat menetralisir sitokin terkait. Penelitian kali ini bertujuan untuk mengetahui pengaruh pemberian vaksin kinoid IL-17A terhadap kerentanan infeksi bakteri pada organ paru mencit model lupus eritematosus sistemik pasca injeksi MRSA (Methicillin resistant staphylococcus aureus).Studi eksperimental menggunakan post test only group design dilakukan terhadap hewan coba mencit balb/c jantan. Sampel dipilih dengan cara proporsional sampling untuk dibagi dalam empat kelompok yaitu kelompok perlakuan 1 (vaksin (+) MRSA (+)) ; kelompok perlakuan 2 (vaksin (+) MRSA (-)) ; kelompok perlakuan 3 (vaksin (-) MRSA (+)) dan kelompok perlakuan 4 (vaksin (-) MRSA (-)). Variabel yang diukur adalah jumlah kolonisasi bakteri pada organ paru pasca injeksi MRSA. Hasil penelitian menunjukkan bahwa terdapat perbedaan jumlah kolonisasi bakteri pada organ paru mencit (Anova, p=0.028). Terjadi perubahan yang signifikan antara kelompok perlakuan 2 dengan kelompok perlakuan 4 (Post hoc tukey, p<0.05). Namun, tidak terbukti terdapat perbedaan yang signifikan antara kelompok perlakuan 2 dengan kelompok perlakuan 1 maupun kelompok perlakuan 3 (Post hoc tukey, p>0.05). Kesimpulan dari penelitian ini adalah vaksin kinoid IL-17A menurunkan jumlah kolonisasi bakteri komensal Staphylooccus aureus pada organ paru mencit balb/c model LES namun belum mampu menurunkan kerentanan infeksi organ paru mencit balb/c model LES terhadap infeksi bakteri MRSA.
English Abstract
Systemic Lupus Erytemathosus is an autoimmune disease which is dangerous. A lot of therapy have been developed to prevent the progrecivity of this disease. One of those theray is kinoid vaccination. Kinoid vaccination is a vaccine which induce the antibody that can neutralized related cytokine. The purpose of this research is to acknowledge the effects of IL-17A vaccination against the bacterial infection susceptibilty on the lungs of lupus mice post injection MRSA (Methicillin Resistant Staphylococcus aureus). Experimental study used post test only group design is being held to the male mice. The samples were selected by random sampling and divided into four groups, namely group 1 (vaccine (+) MRSA (+)) ; group 2 (vaccine (+) MRSA (-)) ; group 3 (vaccine (-) MRSA (+)) ; group 4 (vaccine (-) MRSA (-)).The results showed that there were significant differences in the amount of bacterial colonization on the lungs of mice levels between groups with p value = 0.028 (p <0.05) (One-Way ANOVA test). There was a significant difference between the group 2 and 4. Meanwhile, the group 2 and 1 or 3 showed that there were no significant difference among those groups (Post hoc tukey , p>0.05). The conclusion of ths study is that vaccination of IL-17-A can reduce the number of bacterial colonization of the commensal bacterial Staphylococcus aureus on the lungs of balb/c mice. But, the vaccination can’t decrease the MRSA infection susceptibility of lungs of SLE balb/c mice.
