Abstract

Hipertensi merupakan penyakit kronis tanpa gejala sebelum terjadi komplikasi. Hal ini dapat terjadi akibat disfungsi endotel yang dipengaruhi oleh peningkatan Reactive Oxygen Species (ROS) sehingga menyebabkan peroksidasi lipid yang menghasilkan produk MDA-LDL.Outer Membrane Protein (OMP) Porphyromonas gingivalis memiliki protein gingipain yang memiliki kemiripan dengan MDA-LDL sehingga dapat menginduksi antibodi yang dapat menghambat disfungsi endotel dan proliferasi Vascular Smooth Muscle Cell (VSMC). Penelitian ini bertujuan untuk membuktikan pengaruh pemberian vaksin berbasis OMP bakteri P. gingivalis terhadap proliferasi VSMC pada tikus model hipertensi. Penelitian ini menggunakan 25 tikus wistar (Rattus norvegicus) jantan yang dibagi menjadi 5 kelompok yaitu kontrol positif, kontrol negatif, perlakuan 1 (OMP 100μl), perlakuan 2 (OMP+CFA/IFA 100μl), perlakuan 3 (CFA/IFA 100μl). Pada awal penelitian, ketiga kelompok perlakuan tikus diinjeksikan OMP secara intraperitoneal sesuai dengan ketentuan. Setelah 2 minggu kemudian dilakukan pemberian garam DOCA secara subkutan yaitu 10 mg/kgBB 2 kali seminggu selama 5 minggu. Pemberian booster diberikan sebanyak 3 kali setiap 10 hari secara intraperitoneal. Selanjutnya dilakukan pengukuran tekanan darah tikus dan dilakukan pembedahan untuk mengukur proliferasi VSMC. Hasil analisis dengan uji Kruskal Wallis menunjukkan penurunan proliferasi VSMC (p=0,002) pada tikus yang diberi OMP dan OMP + CFA/IFA. Berdasarkan analisis post Hoc Mann Whitney pada tikus yang diberi OMP dan OMP + CFA/IFA tidak didapatkan perbedaan yang signifikan (p=0.310). Dari hasil penelitian ini dapat disimpulkan bahwa pemberian OMP P. gingivalis dan OMP P.gingivalis + CFA/IFA dapat menurunkan proliferasi VSMC pada tikus model hipertensi, namun perbandingan antara kedua perlakuan tersebut tidak menunjukkan perbedaan yang bermakna secara statistik.

English Abstract

Hypertension is a chronic disease without symptoms before complication happens. This could be an effect of endotel disfunction that affected by increased in Reactive Oxygen Species (ROS) caused lipid peroxidation which produced MDA-LDL. Outer Membrane Protein (OMP) Porphyromonas gingivalis had gingipain protein similar to MDA-LDL that induced antibody which could block the endotel disfunction and Vascular Smooth Muscle Cell (VSMC). This research aimed to prove the influence of OMP bacteria P. gingivalis given towards VSMC proliferation in hypertension rat model. This research use 25 male wistar mice (Rattus norvegicus) divided by 5 group as positive control, negative control, treatment 1 (OMP 100μl), treatment 2 (OMP+CFA+l/IFA 100μl), treatment 3 (CFA/IFA 100μl). At the beginning of the research, 3 groups of mice treatment were injected by OMP intraperitoneal way according to the rules. 2 weeks later they were given DOCA salt subcutaneously 10 mg/lbs twice a week for 5 weeks. OMP repetition was given during 3 times every 10 days by intraperitoneal way. After that, mice blood pressure was measured and surgery performed to measure VSMC proliferation. The Analysis results using Kruskal Wallis test showed reduction VSMC proliferation (p=0,002) in mice that was given OMP P.gingivalis and P.gingivalis + CFA/IFA. The analysis result of Post Hoc Mann Whitney in mice that was given OMP P.gingivalis and P.gingivalis + CFA/IFA is not significant (p=0.310). Based on the result of the research, it can be concluded that OMP P.gingivalis and OMP P.gingivalis + CFA/IFA injections could reduce the VSMC proliferation of hypertension rat model, but the comparison to the treatment did not show a significant differences statistically
