Pengaruh Ekstrak Kacang Tunggak (Vigna Unguiculata) Terhadap Ketebalan Endometrium Tikus Galur Wistar (Rattus Norvegicus) Dengan Ovariektomi

Abstract

Menopause merupakan kondisi wanita yang telah mengalami hipoestrogen. Hormon estrogen berperan penting dalam siklus perkembangan endometrium. Pada kondisi hipestrogen resiko terjadinya perdarahan abnormal uterus akan semakin meningkat. Kacang tunggak (Vigna unguiculata) sebagai fitoestrogen mengandung genistein yang dapat menjadi pilihan alternatif pengganti hormon estrogen. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak kacang tunggak (Vigna unguiculata) terhadap ketebalan endometrium tikus galur wistar (Rattus norvegicus) dengan ovariektomi sehingga hipoestrogen selama 30 hari. Desain penelitian yang digunakan adalah eksperimental dengan rancangan post test only control group design pada 30 ekor tikus dibagi menjadi 5 kelompok, yaitu : kelompok kontrol negatif (tanpa perlakuan), kelompok kontrol positif (dengan ovariektomi), dan 3 kelompok perlakuan dengan ovariektomi serta diberi ekstrak kacang tunggak dengan dosis 1,25 mg/kgBB/hari, 2,5 mg/kgBB/hari, 5 mg/kgBB/hari. Kemudian dilakukan penghitungan ketebalan dengan menggunakan dot slide mikroskop pencahayaan Olympus XC 10 dan software Olyvia dengan perbesaran 400x, dan satuan ketebalan endometrium adalah μm. Analisis One Way ANOVA menunjukkan bahwa pemberian ekstrak kacang tunggak berpengaruh terhadap ketebalan endometrium secara signifikan (p < 0,05) dibanding kelompok kontrol positif. Hasil uji lanjutan Post Hoc LSD didapatkan hasil ketebalan endometrium meningkat dengan pemberian ekstrak kacang tunggak (Vigna unguiculata) dosis 1,25 mg/kgBB/hari dan dosis 2,5 mg/kgBB/hari. Ketebalan endometrium tertinggi terdapat pada pemberian ekstrak kacang tunggak (Vigna unguiculata) dosis 2,5 mg/kgBB/hari. Analisis korelasi Pearson menunjukkan bahwa pemberian dosis ekstrak kacang tunggak (Vigna unguiculata) memiliki korelasi negatif sehingga semakin tinggi dosis akan semakin menurunkan ketebalan endometrium. Kesimpulan dari penelitian ini adalah ekstrak kacang tunggak (Vigna unguiculata) dapat meningkatkan ketebalan endometrium tikus galur wistar (Rattus norvegicus) dengan ovariektomi dengan dosis signifikan 2,5 mg/kgBB/hari.

English Abstract

Menopause is a condition when a woman has exprerienced hipoestrogen period. Estrogen hormone is crucial in endometrial development cyle. In hipoestrogen period, the occurance risk of abnormal uterus bleeding is increasing. Cowpea (Vigna unguiculata) as fitoestrogen contains genistein that can be used as an alternative for estrogen hormone substitute. The aim of this observation is to know the effect of cowpea (Vigna unguiculata) extract towards endometrium thickness in wistar strain rats (Rattus norvegicus) with Ovariectomy so it will reach hipoestrogen in 30 days. The design of this study is experimental using post test only control group design on 30 rats divided into 5 groups, which is: negative control group (without treatment), positive control group (with ovariectomy), and 3 groups with ovariectomy with different dose of cowpea (Vigna unguiculata) extract which is 1.25 mg/kgBW/day, 2.5 mg/kgBW/day, and 5 mg/kgBW/day. Endometrium thickness is then measured using dot slide light microscope Olympus XC 10 and Olyvia software with 400x magnification, using μm as measurement unit of endometrium thickness. One Way ANOVA analysis shows that treatment using cowpea (Vigna unguiculata) extract affects endometrium thickness significantly (p < 0.05) compared to positive control group. Advance test result using Post Hoc LSD method shows that endometrium thickness increases after injected with cowpea (Vigna unguiculata) extract with a dose of 1.25 mg/kgBW/day. The most significant effect is achieved using a dose of 2.5 mg/kgBW/day of cowpea (Vigna unguiculata) extract. Pearson Correlation Analysis shows that treatment using cowpea (Vigna unguiculata) extract has negative correlation, thus the higher the dose of cowpea (Vigna unguiculata) extract is used, the lower the endometrial thickness gets. The conclusion of this observation is that treatment using cowpea (Vigna unguiculata) extract increases endometrium thickness in wistar strain rats (Rattus norvegicus) with ovariectomy with significant dose of 2.5 mg/kgBW/day.
