Abstract

Rimpang temu giring (Curcuma heyneana) mengandung zat aktif kurkumin dan flavonoid yang memiliki aktivitas antioksidan tinggi. Penggunaan gel topikal temu giring mampu menghambat penggelapan pada kulit melalui mekanisme antioksidan. SMEDDS dipilih karena dapat membantu absorbsi senyawa aktif kurkumin dan flavonoid hingga kedalam dermis, menghasilkan sediaan transparan dan stabil terhadap perubahan suhu. Tujuan penelitian ini adalah untuk mengetahui karakteristik dan menguji stabilitas sediaan gel SMEDDS ekstrak etanol 96% temu giring. Dari uji identifikasi kandungan flavonoid dan kurkumin ekstrak etanol 96% temu giring diperoleh hasil positif berupa perubahan warna cokelat pada uji flavonoid dan nilai Rf ekstrak yang sama dengan standar pada uji KLT kurkumin. Sediaan gel SMEDDS ekstrak etanol 96% temu giring dibuat dengan berbagai rasio surfaktan tween 80 dengan span 80, yaitu 60:40, 70:30 dan 80:20. Kemudian dilakukan karakterisasi yang meliputi, pengamatan organoleptis, daya sebar dan homogenitas. Serta uji stabilitas yang meliputi, uji stabilitas suhu kamar selama 6 minggu, uji sentrifugasi dan uji freeze thaw sebanyak 6 siklus. Spesifikasi yang ditetapkan untuk ketiga gel adalah gel berwarna kuning transparan, daya sebar 5,6-6,4 cm dan homogen. Berdasarkan hasil penelitian, dipilih gel dengan rasio surfaktan 80:20 yang menghasilkan karakteristik sesuai spesifikasi dan stabil secara fisik. Gel dengan rasio surfaktan 80:20 menghasilkan sediaan dengan warna kuning transparan berbau mawar dan daya sebar 6,0 ± 0,11 cm. Gel stabil pada penyimpanan suhu kamar selama 6 minggu tanpa paparan cahaya. Stabil pada uji sentrifugasi namun fase minyak dengan air memisah pada uji stabilitas uji freeze thaw.

English Abstract

Temu giring (Curcuma heyneana) rhizome contains active substances curcumin and flavonoid that occupy high antioxidant activity. The effect of the tanning process could be inhibited by using this topical gel through its antioxidant mechanisms. SMEDDS formula was chosen because it could help the absorbtion of curcumin and flavonoid to the dermis. It could also produce clear and thermodinamically stable product. The purpose of this study was to determine the characteristic and the physical stability of temu giring (Curcuma heyneana) rhizome 96% ethanolic extract in SMEDDS gel. The extract was examined for the flavonoid and curcumin content. The results showed that extract’s colour turned to brown for flavonoid test and typical Rf value for extract and curcumin standard (Rf = 0.62). Temu giring rhizome 96% ethanolic extract in SMEDDS gel were made in various surfactant ratios between tween 80 and span 80, which were 60:40, 70:30 and 80:20 respectively. The characteristics of the formulations were determined in terms of visually, spreadability and homogenity. Meanwhile the physical stabilities were tested in the room temperature for 6 weeks, centrifugation test and 6 cycles freeze thaw test. The specifications set were the homogenity and the clearance of the system. The minimum and maximum spreadability were 5.6 and 6.4 respectively. The results showed that the gel formulation with 80:20 surfactant ratio produced the appropriate specifications and physically stable. Gel with 80:20 surfactant ratio produced preparations with clear system and the scent of rose, 6.0 ± 0.11 cm spreadability, stable at room temperature for 2 weeks with light exposure and stable for 6 weeks at room temperature without light exposure. The gel was also stable at centrifugation test, however showed separation phase between oil and water after freeze thaw test.
