Abstract
Latar Belakang: Pada diabetes melitus tipe 2 terjadi peningkatan stres oksidatif sehingga menurunkan enzim antioksidan katalase. Tanaman obat jintan hitam yang digunakan pada terapi penyakit ini memiliki aktivitas antidiabetes dan antioksidan. Namun tanaman obat memiliki kelemahan dalam hal absorbsi sehingga perlu dibuat suatu sistem penghantaran salah satunya dengan nanopartikel PLGA. Tujuan: Mengetahui perbandingan efek nanopartikel PLGA ekstrak biji jintan hitam 48 mg/kg BB, ekstrak biji jintan hitam 48 mg/kg BB, glibenklamid 0,45 mg/kg BB, dan diet normal terhadap kadar katalase pankreas tikus model diabetes melitus tipe 2. Metode: Tikus Sprague dawley 24 ekor dibagi ke dalam 4 kelompok perlakuan lalu diberikan terapi per hari secara sonde intragastrik selama 26 hari. Induksi diabetes melitus tipe 2 dilakukan dengan pemberian diet tinggi lemak selama 40 hari dan injeksi tunggal streptozocin (35 mg/kg BB) secara intraperitoneal. Hasil: Terdapat perbedaan kadar katalase yang signifikan secara statistik (p < 0,05) antara nanopartikel PLGA ekstrak biji jintan hitam dengan non-nanopartikel ekstrak biji jintan hitam (p = 0,000), nanopartikel PLGA ekstrak biji jintan hitam dengan glibenklamid (p = 0,005), dan nanopartikel PLGA ekstrak biji jintan hitam dengan diet normal (p = 0,000). Namun, tidak terjadi penurunan kadar glukosa darah menjadi normal. Kesimpulan: Nanopartikel PLGA ekstrak biji jintan hitam memiliki efek antioksidan pada diabetes melitus tipe 2 dan menunjukkan hasil terbaik dalam meningkatkan kadar katalase pankreas. Aktivitas ini dikarenakan adanya kandungan thymoquinone dalam ekstrak.
English Abstract
Introduction: Type 2 diabetes mellitus is characterized by low level of antioxidant enzyme catalase result from increasing oxidative stress. Nigella sativa is used traditionally to treat and to prevent the complications of type 2 diabetes mellitus through its antidiabetic and antioxidant activities. However, herbal medicines demonstrate poor efficacy due to their poor lipid solubility thus they require drug delivery system, for example PLGA nanoparticle. Objectives: The aim of this study were to determine the effects of 48 mg/kg BW of PLGA loaded nanoparticles of Nigella sativa seed extract on the catalase level in the pancreas tissues of type 2 diabetes mellitus rats compared to those with non-nanoparticles treatment groups, glibenclamide 0.45 mg/kg BW, and normal diet. Methods: Twenty four Sprague dawley rats were randomly assigned into 4 groups which were administered via intragastric per day for 26 days. Type 2 diabetes mellitus were induced by high-fat diet for 40 days and a single intraperitoneally injection of 35 mg/kg BW streptozocin. Results: There were significant differences in the level of catalase (p < 0.05) between PLGA loaded nanoparticles of Nigella sativa seed extract and non-nanoparticle Nigella sativa seed extract (p = 0.000), PLGA loaded nanoparticles of Nigella sativa seed extract and glibenclamide (p = 0.005), and PLGA loaded nanoparticles of Nigella sativa seed extract and normal diet (p = 0.000). However, the plasma glucose level did not decreased to become normal. Conclusion: PLGA loaded nanoparticles of Nigella sativa seed extract had antioxidant effect in type 2 diabetes mellitus and showed the highest pancreatic catalase level. This activity is due to thymoquinone content in the extract.
