Abstract

Aterosklerosis adalah suatu penyakit gangguan metabolisme lemak yang dikarakterisasi oleh deposisi lipoprotein pada dinding arteri dan merupakan respon keradangan kronik sehingga menyebabkan pembentukan plak. Oxidized low density lipoprotein (OxLDL) berperan besar dalam perkembangan aterosklerosis. Lectin-like oxidized low density lipoprotein receptor 1 (LOX-1) merupakan reseptor utama dari OxLDL di endotel pembuluh darah. Penelitian ini bertujuan menentukan pengaruh protein LOX-1 terhadap peningkatan ketebalan dinding aorta tikus Wistar yang diberi diet aterogenik. Penelitian ini menggunakan desain eksperimen murni yang dilakukan 56 hari terhadap 28 tikus Wistar jantan. Tikus dibagi dalam 7 kelompok penelitian yaitu kontrol negatif yang diberi diet normal, kontrol positif yang diberi diet aterogenik, kelompok perlakuan yang diberi aterogenik dan protein LOX-1 dengan dosis 1 ng + alum (P1), 10 ng + alum (P2), 100 ng + alum (P3), 1 μg + alum (P4), dan alum saja (P5). Pada hari ke-57, tikus dieutanasia dan diambil jaringan aorta untuk ditentukan ketebalan dinding aortanya menggunakan pengecatan Hematoksilin Eosin (HE). Analisis dengan uji One-Way ANOVA menunjukkan bahwa pemberian vaksin mampu mencegah penebalan dinding aorta secara bermakna pada kelompok perlakuan dibandingkan dengan kelompok kontrol positif (p < 0,05), dan tidak ada perbedaan bermakna antara kelompok perlakuan dengan kelompok kontrol negatif (p > 0,05). Terdapat hubungan yang berlawanan antara pemberian dosis protein LOX-1 dengan ketebalan dinding aorta, semakin besar dosis protein LOX-1 maka semakin besar potensinya dalam menghambat peningkatan penebalan dinding aorta (p = 0,053; r = - 0,377; Uji Pearson). Kesimpulan dari penelitian ini adalah protein LOX-1 berpotensi dalam mencegah progresivitas plak aterosklerosis melalui penghambatan peningkatan ketebalan dinding aorta tikus Wistar dengan diet aterogenik.

English Abstract

Atherosclerosis is a lipid metabolism disorder which is characterized by the lipoprotein deposition in the artery wall. It is associated with a chronic inflammatory response which causes the atherosclerotic plaques formation. Oxidized low density lipoprotein (OxLDL) plays an important role on atherosclerosis development. Lectinlike oxidized low density lipoprotein receptor 1 (LOX-1) is the primary receptor for OxLDL in the endothelial cells. The aim of this study is to determine the potency of LOX-1 protein to prevent aortic wall thickness in Wistar rats which were received atherogenic diet. This study used true experimental design for 56 days with 28 male Wistar rats. Rats were divided into 7 groups i.e. negative control with normal diet, positive control with atherogenic diet, treatment group with LOX-1 protein 1 ng + alum (P1), 10 ng + alum (P2), 100 ng + alum (P3), 1μg + alum (P4) and alum only (P5). On the 57th day, the rats were sacrificed and the aorta tissues were taken to be determine aortic wall thickness with Hematoxylin Eosin (HE) staining. One-Way ANOVA showed that the administration of LOX-1 protein could significantly inhibit aortic wall thickening in the treatment groups compared with the positive control group (p < 0,05), and there is no significant differences between the treatment groups with the positive control group (p > 0,05). There is a contradictory relationship between the treatment groups with the aortic wall thickness which is the higher the dose of LOX-1 protein, the higher its potency to prevent the aortic wall thickening (p = 0,053; r = - 0,377; Pearson test). The conclusion of this study is the LOX-1 protein has the potency to prevent the progression of atherosclerotic plaque by inhibiting the aortic wall thickening in Wistar rats with atherogenic diet.
