Abstract
Aterosklerosis adalah penyakit degeneratif kronis pada pembuluh darah yang dipicu oleh karena pembentukan plak pada dinding arteri yang merupakan penyebab utama kegawatdaruratan penyakit kardiovaskuler, meliputi jantung serta pembuluh darah dan merupakan penyebab pertama kematian secara global. Diet aterogenik memicu terjadinya respon keradangan yang ditandai oleh peningkatan jumlah sel T CD4+. Lectin-Like Oxidized LDL Receptor 1 (LOX-1) merupakan reseptor Ox-LDL pada sel endotel yang berperan penting dalam patogenesis aterosklerosis. Penelitian ini bertujuan menentukan pengaruh pemberian protein LOX-1 terhadap jumlah sel T CD4+ pada tikus Wistar yang diberi diet aterogenik. Penelitian ini merupakan penelitian eksperimental laboratorik menggunakan postest only control group design yang dilakukan 56 hari terhadap 28 tikus Wistar jantan. Tikus dibagi dalam 7 kelompok penelitian yaitu kontrol negatif yang diberi diet normal, kontrol positif yang diberi diet aterogenik, kelompok perlakuan yang diberi diet aterogenik dan protein LOX-1 dengan dosis 1 ng + alum (P1),10 ng + alum (P2), 100 ng + alum (P3), 1 µg + alum (P4), dan alum saja (P5). Pada akhir penelitian, jaringan limpa pada tikus diambil untuk menentukan jumlah sel T CD4+ dengan metode immunohistokimia. Analisis dengan uji One-Way ANOVA didapatkan nilai signifikansi p<0,05. Kesimpulan dari penelitian ini adalah pemberian protein LOX-1 belum dapat meningkatkan jumlah sel T CD4+. Peningkatan jumlah sel T CD4+ yang paling tinggi didapatkan pada kelompok P5 dengan pemberian alum saja.
English Abstract
Atherosclerosis is a chronic degenerative disease of blood vessels which lead to the plaque formation in the artery wall as main cause of emergency in cardiovascular disease include heart and blood vessels as the first cause of death globally. Atherogenic diet triggers inflammatory response characterized by increasing the number of T CD4+ cells number. Lectin-Like Oxidized LDL Receptor 1 (LOX-1) is a scavanger receptor of Ox-LDL on endothelial cells that play an important role in the pathogenesis of atherosclerosis. The aim of this study is to determine the effect of LOX-1 admission protein to T CD4+ cells numbers in the spleen of Wistar rats fed with atherogenic diet. This study was experimental laboratory with postest only control group design for 56 days using 28 male Wistar rats. Rats were divided into 7 groups which were negative control with normal diet, positive control with atherogenic diet, treatment group with LOX-1 protein 1 ng + alum (P1), 10 ng + alum (P2), 100 ng + alum (P3), 1 µg + alum (P4) and alum only (P5). At the end of the study, spleen tissues were taken to determine its T CD4+ cells number with immunohistochemical method. One-Way ANOVA results test showed significancy level p<0.05. The conclusion of this study is the admission of LOX-1 protein have not be able to increase the number of T CD4+ cells. The highest increasing number of the T CD4+ cells on treatment group is P5 group with alum only.
