Abstract

Kadmium merupakan salah satu limbah aktivitas pertambangan batu bara yang mencemari air Sungai Mahakam dengan kadar 0,06 mg/L. Konsumsi air Sungai Mahakam diduga berkaitan dengan kasus dismenore primer. IL-1β meru¬pakan sitokin proinflamasi yang produksinya dapat diinduksi kadmium untuk me¬ningkatkan secara abnormal produksi PGF2α melalui lisis membran fosfolipid, sintesis COX-2, dan sintesis AKR1B1. PGF2α merupakan biomarker dismenore primer. Penelitian dilakukan dengan tujuan untuk mengetahui apakah pemberian kadmium per oral meningkatkan ekspresi sitokin IL-1β stroma endometrium. Studi eksperimental dilakukan dengan post test only control group design dengan menggunakan subjek hewan coba tikus Wistar betina. Sampel dipilih mengguna¬kan simple random sampling menjadi 4 kelompok, yaitu kelompok kontrol nega¬tive (KN) (tanpa pemberian kadmium) dan 3 kelompok perlakuan (KP1, KP2, dan KP3) dengan dosis kadmium yang disondekan berturut-turut 0,03 µg/kgBB/L/hari, 0,06 µg/kgBB/L/hari, dan 0,09 µg/kgBB/L/hari. Variabel yang diukur adalah jum¬lah ekspresi IL-1β intrasel yang dipresentasikan dengan intensitas resiprokal ha¬sil pengecatan IHK pada 2 subjek KN, 4 subjek KP1, 3 subjek KP2, dan 3 subjek KP3. Hasil penelitian dan analisa data menunjukkan bahwa pemberian kadmium pada kelompok perlakuan tidak meningkatkan secara signifikan ekspresi sitokin IL-1β stroma endometrium subjek (p-value uji Kruskall Wallis=0,12) dibandingkan kelompok kontrol negatif. Berdasarkan penelitian ini, dapat disimpulkan bahwa pemberian kadmium tidak memberikan peningkatan ekspresi sitokin IL-1β stroma endometrium secara signifikan. Peneliti menyarankan untuk dilakukannya peneli-tian mengeksplorasi faktor-faktor lain yang berperan menghambat sitotoksisitas kadmium, produksi IL-1β, dan meningkatkan PGF2α.

English Abstract

Cadmium is one of the coal mining activity wastes that pollutes the Mahakam River water with the levels of 0.06 mg/L. Resident’s Mahakam River water consumption is related to frequent cases of primary dysmenorrhea. IL-1β is a proinflammatory cytokine of which production may be induced cadmium may abnormally increase the PGF2α production via lysis of membrane phospholipids, synthesis of COX-2, and synthesis AKR1B1. PGF2α is a biomarker of primary dysmenorrhea. The study was conducted in order to determine whether oral administration of cadmium increased the expression of endometrial stroma IL-1β cytokine. Experimental studies was conducted with post-test only control group design using the subject of female Wistar rats. The sample was selected using a simple random sampling into 4 groups, namely the negative control group (NCG) (without giving cadmium) and 3 treatment groups (TG1, TG2 and TG3) at consecutively given orogastrically cadmium doses of 0.03 µg/kg of BW/L/day, 0.06 µg/kg of BW/L/day, and 0.09 µg/kg of BW/L/day. The measured variable was the amount of intracellular IL-1β cytokines expression presented with the results of IHC staining reciprocal intensity at 2 subjects of NCG, 4 subjects of TG1, 3 subjects of TG2, and 3 subjects of TG3. The research and analysis of data showed that the administration of cadmium in the treatment group did not increase the endometrial stroma IL-1β cytokines expression significantly (p-value Kruskal Wallis test = 0.12) of treated group compared to the negative control group. Based on this study, it can be concluded that cadmium administration did not provide an increase in the expression of cytokines IL-1β significantly endometrial stroma. Researcher suggest to undertake research to explore other factors that inhibit the cytotoxicity of cadmium, the production of IL-1β, and increased PGF2α.
