Abstract
Vegfr-2 Adalah Reseptor Yang Menengahi Tindakan Pertumbuhan Dan Permeabilitas Vegf. Bromelain Dikatakan Memiliki Efek Menghambat Angiogenesis. Embrio Ayam Digunakan Karena Perkembangan Yang Cepat Dan Lebih Murah Dibandingkan Mamalia (Tikus) Karena Studi Tidak Memungkinkan Dilakukan Pada Manusia. Penelitian Ini Bertujuan Untuk Mengetahui Pengaruh Pemberian Bromelain Berbagai Dosis Terhadap Penurunan Ekspresi Vegfr-2. Penelitian Ini Didesain Sebagai Suatu Randomized Control Group Post Test Study. Telur Diinkubasi Selama Minimal 16 Jam Dalam Keadaan Horizontal Pada Suhu 15-20º C. Telur Kemudian Dikelompokkan Menjadi 4 Kelompok Yaitu A (Pemberian Bromelain 0 µg), B (Pemberian Bromelain 50 µg), C (Pemberian Bromelain 100 µg) Dan D (Pemberian Bromelain 200 µg). Setelah Inkubasi Dalam Inkubator Dengan Suhu 37,5-38,5º C Selama 48 Jam Telur Dipecah Dan Diamati. Selanjutnya Dilakukan Pengecatan Imunohistokimia Dan Menghitung Ekspresi Vegfr-2 Dengan Menggunakan Unbiased Sterology. Uji Statistik Anova Menunjukkan Adanya Penurunan Yang Signifikan Pada Setiap Dosis Yang Diberikan. Hasil Dari Penelitian Ini, Pemberian Bromelain Menurunkan Ekpresi Vegfr-2. Pada Penelitian Yang Lebih Lanjut Disarankan Menggunakan Parameter Gambaran Histologis Pembuluh Darah Agar Lebih Jelas
English Abstract
Vegfr-2 Is A Receptor That Mediates The Actions Of Vegf Growth And Permeability. Bromelain Is Known Of Angiogenesis Inhibitor. The Chick Embryo Has For Long Been One Of The Most Widely Used Laboratory Animals For Research And The Reasons Are Clear To See Fertilized Eggs Are Cheap And Its Development Is Fast In Comparison With Most Mammalian Embryos And Similar To That Of The Mouse. This Study Aimed To Determine The Effect Of Various Doses Of Bromelain To Decrease The Expression Of Vegfr-2. This Is Purely An Experimental Study With Randomized Control Group Post Test Design. The Fertilized Chicken Eggs Were Incubated For A Minimum Of 16 Hours In A Horizontal State At A Temperature Of 15-20º C. They Were Divided Into 4 Group: A (0 µg Bromelain Administration), B (50 µg Bromelain Administration), C (100 µg Bromelain Administration) And D (200 µg Bromelain Administration). After Incubation In An Incubator At A Temperature Of 37.5 To 38.5º C For 48 Hours, Eggs Were Broke And Observed. Eggs Were Stained By Immunohistochemistry And Vegfr-2 Expression Were Counted By Using Unbiased Sterology. Statistics Anova Showed A Significant Decrease Vegfr-2 Expression In Any Given Dose. The Result Of This Study, Administration Of Bromelain Lowered Expression Of Vegfr-2. Further Research Recommended To Use Histological Of Blood Vessels Parameter.
