Abstract

Dislipidemia merupakan gangguan metabolisme lipoprotein yang salah satunya ditandai oleh peningkatan kadar kolesterol total di dalam darah. Kacang tunggak merupakan suatu bahan makanan yang mengandung isoflavon yang dapat menghambat aktivitas enzim HMG-KoA reduktase dalam proses kolesterologenesis di hati. Antioksidan yang terkandung dalam kacang tunggak mampu menurunkan absorpsi kolesterol di dalam usus. Tujuan dari penelitian ini adalah untuk mengetahui pemberian yoghurt kacang tunggak dapat mencegah peningkatan kadar kolesterol total tikus wistar yang diberi diet tinggi lemak. Penelitian ini menggunakan studi eksperimental dengan desain penelitian Post test Control Group yang dilakukan pada 30 ekor tikus jantan dan dibagi menjadi 5 kelompok yaitu: kelompok kontrol negatif diberikan diet normal, kelompok kontrol positif diberikan diet tinggi lemak, kelompok perlakuan 1 diberikan diet tinggi lemak dan yoghurt kacang tunggak dosis isoflavon 1,6 mM / g BB, kelompok perlakuan 2 diberikan diet tinggi lemak dan yoghurt kacang tunggak dosis isoflavon 2,4 mM / g BB serta kelompok perlakuan 3 diberikan diet tinggi lemak dan yoghurt kacang tunggak dosis isoflavon 3,2 mM/ g BB. Perlakuan dilakukan selama 8 minggu. Hasil penelitian menunjukkan bahwa kadar kolesterol total antara kelima kelompok berbeda signifikan (Kruskal-Wallis, p = 0.000). Kesimpulan penelitian ini adalah pemberian yoghurt kacang tunggak dapat mencegah peningkatan kadar kolesterol total serum pada tikus wistar yang diberi diet tinggi lemak.

English Abstract

Dyslipidemia is lipoprotein metabolism disorder which is marked by an increasing of total cholesterol level in blood serum. Cowpea contains a lot of isoflavone that can inhibit the activity of the enzyme HMG-CoA reductase in kolesterologenesys process. Antioxidant in cowpea can be able to lower cholesterol absorption by the intestine. The aims of this study is to find out cowpea yogurt which can prevent the increasing total cholesterol level of wistar rat induced by high-fat diet. This study used an experimental study with post test only control group design in 30 male rats. It was devided into 5 groups: negative control group was given normal diet, positive control group was given high-fat diet, treatment group 1 was given high-fat diet and cowpea yogurt isoflavone dose 1,6 mM / g BW, treatment group 2 was given high-fat diet and cowpea yogurt isoflavone dose 2,4 mM / g BW as well as treatment group 3 was given high-fat diet and cowpea yogurt isoflavone dose 3,2 mM / g BW. The treatment was done for 8 weeks. Result showed that total cholesterol level among fifth group was significant difference (Kruskal-wallis, p = 0,000). The conclusion of this study is cowpea yogurt can prevent the increasing total cholesterol levels of wistar rats induced by high-fat diet.
