Abstract

Keamanan konsumsi Genistein pada kehamilan trimester pertama dan pengaruhnya pada perkembangan embrio, terutama pada perkembangan sistem saraf pusat masih belum banyak diketahui. Penelitian ini bertujuan untuk mengetahui pengaruh paparan genistein terhadap perkembangan awal sistem saraf pusat. Model yang digunakan dalam penelitian ini adalah embrio ayam (Gallus gallus) umur 48 jam. Genistein pada dosis 5 µM, 10 µM dan 20 µM diinjeksikan secara in ovo ke dalam yolk sac telur sebelum diinkubasi. Telur kemudian diinkubasi selama 48 jam pada suhu 37,50 – 38,50 C. Setelah 48 jam, cangkang telur dipecah, embrio diambil dan dilakukan pewarnaan menggunakan Toluidine Blue. Evaluasi dilakukan pada neural tube, neuropore anterior, neuropore posterior dan somit. Pada penelitian ini, ditemukan defek neural tube pada kelompok perlakuan dengan genistein lebih banyak dibanding kelompok kontrol namun secara statistik hasil tersebut tidak signifikan sedangkan untuk somit, pada kelompok perlakuan genistein dengan dosis 10 µM, jumlah somit lebih banyak dibanding kelompok kontrol dan secara statistik jumlah tersebut signifikan.

English Abstract

The safety of genistein consumption in the first trimester of pregnancy and its effect on embryonic development, especially in the development of Central Nervous System (CNS) is still not widely known. This study aimed to determine the effect of genistein exposure to the early development of the Central Nervous System. The animal model used in this research is chick embryo (Gallus gallus). Genistein at a dose of 5 µM, 10 µM and 20 µM in ovo injected into the yolk sac of the eggs before incubation. The eggs were then incubated for 48 hours at a temperature of from 37.50 to 38.50 C. After 48 hours, broken egg shell, embryos are taken and carried out using Toluidine Blue staining. The evaluation was done on the neural tube, anterior neuropore, posterior neuropore and somites. In this study, it was found neural tube defects in the group treated with genistein more than the control group but the result was not statistically significant, while for the somites, in the group treated with a dose of 10 µM genistein, the number of somites more than the control group and statistically the number is significant
