Abstract

Shigella dysenteriae merupakan bakteri batang Gram negatif yang bersifat aerob atau fakultatif anaerob dari famili Enterobacteriaceae yang dapat menyebabkan disentri basiler atau penyakit shigellosis. Shigella dysenteriae telah dilaporkan resisten terhadap berbagai macam antibiotik seperti ampisilin, tetrasiklin, streptomisin, dan kloramfenikol. Oleh karena itu, diperlukan alternatif pengobatan dalam mengatasi bakteri Shigella dysenteriae. Salah satunya dengan menggunakan tanaman hias sekaligus obat seperti daun lidah mertua (Sansevieria trifasciata var.Laurentii). Daun lidah mertua memiliki kandungan aktif yang diduga dapat menjadi antibakteri seperti saponin, tanin dan polifenol. Penelitian ini bertujuan untuk membuktikan pengaruh ekstrak etanol daun lidah mertua terhadap Shigella dysenteriae secara in vitro. Penelitian ini merupakan penelitian eksperimental laboratorik dengan menggunakan dilusi tabung untuk menentukan KHM (Kadar Hambat Minimal) dan KBM (Kadar Bunuh Minimal). Sampel yang digunakan dalam penelitian ini adalah isolat Shigella dysenteriae yang diperoleh dari Laboratorium Mikrobiologi Fakultas Kedokteran Universitas Brawijaya, Malang, Jawa Timur. Konsentrasi yang digunakan dalam penelitian yaitu 3.75% v/v, 5% v/v, 6.25% v/v, 7.5% v/v, dan 8.75% v/v dengan empat kali pengulangan, sedangkan konsentrasi Shigella dysenteriae adalah 106CFU/ml. Hasil uji statistik menunjukan bahwa ekstrak etanol daun lidah mertua, secara signifikan dapat menghambat pertumbuhan Shigella dysenteriae (Kruskal Wallis, p<0.05) dan terdapat hubungan antara peningkatan konsentrasi ekstrak etanol daun lidah mertua dengan penurunan jumlah pertumbuhan koloni Shigella dysenteriae (Spearman rho = -0.975, p = 0.000). KHM ekstrak etanol daun lidah mertua terhadap Shigella dysenteriae adalah 7.5% v/v dan KBM-nya adalah 8.75% v/v. Dengan demikian, dapat disimpulkan bahwa ekstrak etanol daun lidah mertua memiliki efek antibakteri terhadap Shigella dysenteriae secara in vitro.

English Abstract

Shigella dysenteriae is a Gram-negative rod bacteria. It is aerobic or facultative anaerobic bacteria of family Enterobacteriaceae which can cause bacillary dysentery or shigellosis disease. Shigella dysenteriae has been reported to be resistant to various kinds of antibiotics such as ampicillin, tetracycline, streptomycin, and chloramphenicol. Therefore, it is necessary to address the alternative treatment of Shigella dysenteriae bacteria. One of them using medicinal plants as well as Sansevieria trifasciata Leaf (Sansevieria trifasciata var.Laurentii). Sansevieria trifasciata Leaf has active compounds supposed to be antibacterial such as saponins, tannins and polyphenols. This study aimed to verify the effect of the ethanol extract of Sansevieria trifasciata Leaf towards Shigella dysenteriae by in vitro. This study was a laboratory experimental study using tube dilution to determine the MIC (Minimum Inhibitory Concentration) and MBC (Minimum Bactericidal Concentration). The sample of which used in/(of) this study is isolates of Shigella dysenteriae obtained from the Laboratory of Microbiology, Faculty of Medicine, University of Brawijaya, Malang, East Java. The concentration used of the study is 3.75% v/v, 5% v/v, 6.25% v/v, 7.5% v/v, dan 8.75% v/v by four times repetitions, while the concentration of Shigella dysenteriae is 106CFU/ml. Statistical test results showed that the ethanol extract of Sansevieria trifasciata Leaf, can significantly inhibit the growth of Shigella dysenteriae (Kruskal-Wallis, p<0.05) and there is a relationship between an increased concentration of the ethanol extract of the leaves of Sansevieria trifasciata Leaf with a decrease in the number of colony growth of Shigella dysenteriae (Spearman rho = -0.975, p = 0.000). MIC ethanol extract of Sansevieria trifasciata Leaf towards Shigella dysenteriae is 7.5% v/v and for MBC is 8.75% v/v. Hence, the conclusion is the ethanol extract of Sansevieria trifasciata Leaf has an antibacterial effect towards Shigella dysenteriae by in vitro.
