Abstract
Neutrofil dari pasien LES memiliki kemampuan lebih dalam memproduksi neutrophile extracellular traps (NETs) terutama sebagai respon terhadap kompleks antibodi. NETs mampu mengeluarkan substansi bakterisidal protein imunostimulasi, dan autoantigen seperti LL-37, IL-17, dan dsDNA. Dengan demikian, NETosis pada jaringan berhubungan dengan peningkatan anti dsDNA pada serum. Penelitian ini bertujuan untuk mengetahui korelasi pembentukan NETs dengan kadar antibodi anti dsDNA pada pasien lupus Eritematosus Sistemik. Pengambilan sampel secara purposive pada 28 pasien LES yang diperoleh dari Poli Rheumatologi bagian Penyakit Dalam dan ruang rawat inap Penyakit Dalam di Rumah Sakit Saiful Anwar, Malang. Seluruh subyek diperiksa kadar pembentukan NETs dan kadar anti dsDNA. Kadar pembentukan NETs diukur melalui peningkatan persentasi absorbansi MPO-DNA yang dikeluarkan oleh neutrofil saat NETosis dibandingkan dengan neutrofil yang tidak diinduksi PMA menggunakan metode ELISA. Kadar antibodi anti dsDNA diukur dengan metode ELISA. Hasil pengamatan dianalisis menggunakan uji korelasi spearman dengan bantuan program SPSS 17.0. Signifikansi statistik ditetapkan jika nilai p < 0,05. Hasil penelitian ini tidak menunjukkan adanya korelasi antara pembentukan NETs yang diukur melalui kadar MPO-DNA dengan kadar antibodi anti dsDNA pada pasien lupus Eritematosus Sistemik (r: -0,127, p: 0,519). Eksternalisasi antigen oleh NETs ikut menyumbang peran dalam terbentuknya antibodi anti dsDNA pada pasien LES namun, mungkin terdapat faktor lain yang lebih dominan dalam mempengaruhi tingginya kadar antibodi anti dsDNA. Kesimpulan dari penelitian ini tidak terdapat korelasi antara pembentukan NETs yang diukur melalui kadar MPO-DNA secara in vitro dengan kadar anti dsDNA pada pasien lupus eritematosus sistemik.
English Abstract
Neutrophils from SLE patients has more capability in producing neutrophile extracellular traps (NETs) primarily as a response to antibody complexes. NETs has capability releasing the substance of bactericidal proteins immunostimulation, and autoantigen such as LL-37, IL-17, and dsDNA. Thus, tissue NETosis associated with an increase anti-dsDNA. This study aims to determine the correlation NETs formation with anti-dsDNA antibody levels in patients with Systemic Lupus Erythematosus. Purposive sampling in 28 patients with SLE obtained from Poly Rheumatology Internal Medicine Department or inpatient room Internal Medicine Department in Saiful Anwar Hospital, Malang. All subjects examined levels of the formation of NETs and anti dsDNA levels. NETs formation levels were measured by an increase in the percentage of absorbance MPO-DNA released by neutrophils when NETosis compared to non-induced neutrophil by PMA using ELISA method. Anti-dsDNA antibody levels were measured by ELISA method. Results were analyzed using Spearman correlation test with SPSS 17.0. Statistical significance was determined if the value of p <0.05. The results of this study showed no correlation between the formation of the NETs as measured by levels of MPO-DNA with anti-dsDNA antibody levels in patients with Systemic Lupus Erythematosus (r: -0.127, p: 0.519). Antigen externalization by NETs contribute a role in the formation of anti-dsDNA antibodies in patients with SLE however, there may be other factors that are more dominant in influencing the high anti-dsDNA antibody levels. In conclusion there is no correlation between the formation of the NETs as measured by levels of MPO-DNA in vitro with anti-dsDNA levels in patients with systemic lupus erythematosus
