Abstract

Di negara berkembang seperti Indonesia, insiden kanker kolon meningkat pada umur diatas 50 tahun. Kanker kolon dapat disebabkan karena inflamasi kronis pada kolon yang disebut Colitis Associated Colon Cancer (CAC). CAC merupakan hasil dari proses inflamasi kronis yang melibatkan sel-sel imun dan sitokinnya. Limpa mempunyai peran dalam patogenesis kolitis, yaitu menghasilkan sel-sel imunitas terhadap patogen atau antigen asing. Ekstrak etanol daun Benalu Mangga (Dendrophthoe pentandra) mengandung senyawa flavonoid jenis kuersetin sebagai anti kanker. Penelitian ini bertujuan untuk mengetahui pemberian ekstrak etanol daun Benalu Mangga (Dendrophthoe pentandra) terhadap penurunan jumlah sel neutrofil pada limpa mencit BALB/C model Colitis-associated Colon Cancer. Studi eksperimental ini menggunakan desain penelitian post test only control group selama 15 minggu di Lab Parasit FKUB. Terdapat 5 kelompok perlakuan yaitu dua kontrol dan tiga perlakuan dosis ekstrak etanol daun Benalu Mangga (125, 250 dan 500 mg/kgBB). Pengamatan histopatologi dilakukan dengan menghitung infiltrasi sel neutrofil pada limpa mencit dengan pewarnaan H&E. Hasil dari statistik one way ANOVA menunjukkan penurunan jumlah sel neutrofil secara signifikan pada kelompok E (p < 0,05) setelah perlakuan dan pemberian ekstrak, dilakukan uji korelasi Pearson yang mempunyai nilai r = - 0,975 yaitu mempunyai korelasi yang kuat antara ekstrak etanol daun Benalu Mangga (Dendrophthoe pentandra) dengan penurunan jumlah sel neutrofil. Dapat disimpulkan bahwa pemberian ekstrak etanol daun Benalu Mangga (Dendrophthoe pentandra) dapat menurunkan jumlah sel neutrofil pada kolon mencit model CAC.

English Abstract

In Indonesia, as one of developing countries, colon cancer phenomenon is increasing at the age over 50s. This cancer can be caused by chronic inflammation in colon, namely, Colitis-Associated Colon Cancer (CAC). CAC is the result of a chronic inflammatory process involving the immune cells and cytokines. The spleen has a role in the pathogenesis of colitis; it produces immune cells against pathogens or foreign antigens. Ethanol extract from leaf of Benalu Mangga (Dendrophthoe pentandra) contains flavonoids, quercetin type, as an anti- cancer. This study aimed to determine the effect of applying ethanol extract of Benalu Mangga leaf toward the decrease of neutrophils in the spleen cells of mice BALB/c model of Colitis associated Colon Cancer. This experimental study applies posttest-only-control-group research design for 15 weeks in Parasite Laboratory, Medical Faculty of UB. There are 5 treatment group i.e. two control groups and 3 treatment groups with ethanol extract from leaf of Benalu Mangga (125, 250 and 500 mg/kgBB). Histopathological observation is investigated by computing the infiltration of neutrophils in the spleen cells of mice stained with H & E. The results of one-way ANOVA statistics significantly show a reduction of neutrophil cell number (p < 0.05). After giving the treatment and applying the extract, the Pearson correlation test, r = - 0.975, is applied. The result shows that it has a strong correlation between ethanol extract of Benalu Mangga leaf with a reduction of neutrophil cell numbers. It can be concluded that the Ethanol extract of Benalu Mangga leaf can reduce the number of neutrophil cells in the colon of mice models of Colitis-Associated Colon Cancer
