Abstract

Sirosis hepar adalah inflamasi kronis pada hepar dengan berbagai macam penyebab. Inflamasi yang terus-menerus terjadi akan menyebabkan terbentuknya jaringan fibrous. Akibatnya hepar tidak berfungsi dengan baik, bahkan hepar akan mati. Penggunaan terapi regeneratif sangat berpotensi untuk dikembangkan. Mengingat pengobatan paling baik saat ini transplantasi, memiliki angka kematian yang tinggi. Beta-glucan yang kita dapatkan dari ekstrak oats meningkatkan kadar G-CSF dalam tubuh. Peningkatan kadar G-CSF dalam tubuh akan melepaskan hematopoietic stem cells dalam sirkulasi tubuh. Hematopoietic stem cells yang telah dilepaskan ke sirkulasi akan meregenerasi hepar yang telah rusak. Tujuan penelitian ini untuk mengetahui peran pemberian ekstrak oats dalam meningkatkan protein total yang merupakan tanda dari meningkatnya fungsi fisiologis dari hepar. Penelitian ini menggunakan desain eksperimental (true experimental design) di laboratorium secara in vivo menggunakan rancangan randomized post test only controlled group design. Tikus wistar sebagai hewan coba dibagi menjadi 5 kelompok, yaitu : kontrol negatif, kontrol positif, Perlakuan 1 (CCL4 + Ekstrak Oats 90mg/kgBB), Perlakuan 2 (CCL4 + Ekstrak Oats 180mg/kgBB), Perlakuan 3 (CCL4 + Ekstrak Oats 360mg/kgBB). CCL4 untuk menginduksi kerusakan hepar diinjeksikan 1ml/kgBB 3 kali seminggu secara subcutaneus selama 8 minggu. Ekstrak diberikan setiap hari mulai minggu ke-3 hingga minggu ke-8. Hasil penelitian menunjukan bahwa pemberian ekstrak oats mampu meningkatkan kadar protein total secara signifikan (p<0,05). Dengan demikian, pemberian ekstrak oats mampu mengembalikan fungsi fisiologis hepar yang rusak.

English Abstract

Cirrhosis of the liver is a chronic inflammation of the liver with a wide variety of causes. Persistent inflammation occurs will lead to the formation of fibrous tissue. As a result, the liver is not functioning properly, and even liver will die. The use of regenerative therapy has the potential to be developed. The current best treatment of transplantation, has a high mortality rate. Beta-glucan extract that we get from oats increase levels of G - CSF in the body. Increased levels of G - CSF in the body will release hematopoietic stem cells in circulation. Hematopoietic stem cells that have been released into circulation will regenerate damaged liver. The purpose of this study was to determine the role of oats extract in increasing total protein in body, which is one of the indicators of the improvement of physiological functions of the liver. This study used an experimental design (true experimental design) in the laboratory in vivo using a design Randomized controlled group post test only design. Wistar rats as experimental animals were divided into 5 groups: negative control , positive control , treatment 1 ( CCl4 + extract Oats 90mg/kgBB ) , Treatment 2 ( CCl4 + extract Oats 180mg/kgBB ) , Treatment 3 ( CCl4 + Oats Extract 360mg / kg ) . CCl4 injected to induce liver damage 1ml/kgBB subcutaneus 3 times a week for 8 weeks. The extract was given daily from week 3 to week 8. The results showed that the extract of oats were able to increase the total protein content significantly ( p < 0.05 ). Thus, oats extract is able to restore physiological functions of the liver which is damaged.
