Abstract
Infeksi Staphylococcus aureus saat ini telah berkembang menjadi masalah kesehatan yang serius pada neonatus, bayi, anak, dan dewasa. Menurut WHO, case fatality rates infeksi Staphylococcus aureus mencapai 30%. Selain itu Staphylococcus aureus juga dapat menyebabkan infeksi pada luka post operasi caesar . Saat ini, beberapa strain Staphylococcus aureus diketahui telah resisten terhadap antibiotik, salah satunya adalah Methicillin-resistant Staphylococcus aureus atau MRSA. Kulit lemon diketahui memiliki beberapa kandungan yang diduga bersifat antibakteri yaitu limonen, kumarin dan flavonoid. Penelitian ini bertujuan untuk mengetahui efektivitas ekstrak kulit lemon terhadap pertumbuhan Staphylococcus aureus secara in vitro . Sampel diperoleh dari isolat Staphylococcus aureus di Laboratorium Mikrobiologi FKUB. Konsentrasi ekstrak yang dipakai yaitu 2,5%, 3%, 3,5%, 4%, 4,5%, 5%, 5,5% dan 0% (KK). Metode penelitian menggunakan dilusi tabung dengan empat kali pengulangan pada setiap konsentrasi. Hasil statistik One-Way ANOVA menunjukkan terdapat perbedaan yang signifikan pada perubahan konsentrasi ekstrak kulit lemon terhadap jumlah koloni Staphylococcus aureus (p<0,05). Uji korelasi dan regresi menunjukkan adanya hubungan yang erat dan berbanding terbalik serta pengaruh yang signifikan antara konsentrasi ekstrak dengan jumlah koloni bakteri (Korelasi, r= -0,827; p<0,05). Berdasarkan penelitian ini, dapat disimpulkan bahwa ekstrak kulit lemon mempunyai efek antibakteri terhadap bakteri Staphylococcus aureus dengan KHM 4% dan KBM 5,5%.
English Abstract
Nowadays Staphylococcus aureus infection has emerged as a serious health problem to neonatus, infant, child and adult. According to WHO, case fatality rates in Staphylococcus aureus infections can reach 30%. Beside that Staphylococcus aureus can cause post caesar wound infections. Recently, some strains of Staphylococcus aureus have been known for their antibiotic resistance such as Methicillin-resistant Staphylococcus aureus , that is widely known as MRSA. Lemon peels contain some substances that are predicted to have antibacterial effects such as limonene, coumarin and flavonoid. This research’s goal is to identify the effectivity of lemon peels extract against the growth of Staphylococcus aureus in vitro. The sample is obtained Staphylococcus aureus isolate from Microbiology Laboratory of Medical Faculty of Brawijaya University. The extract concentrate used is 2,5%, 3%, 3,5%, 4%, 4,5%, 5%, 5,5% and 0% (KK). This research used the Tube Dilution Test as research method with four repetition in each concentration. The result of One-Way ANOVA statistics shows there is a significant difference in the change of lemon peels extract concentration to the amount of the Staphylococcus aureus colony (p<0,05). Correlation-Regression test indicated close and reverse correlation and also significant influence of extract’s concentration against the number of bacterial colony (correlation, r= -0,827; p<0,05). Based on this research it can be concluded that the lemon peels extract has an antibacterial effect against Staphylococcus aureus with MIC 4% and MBC 5,5%.
