Abstract

Staphylococcus aureus pada individu yang sehat sering ditemukan sebagai flora normal di rongga mulut, namun bila terjadi gangguan keseimbangan bakteri ini juga dapat menjadi penyebab infeksi pulpa. Propolis merupakan zat yang dihasilkan oleh lebah untuk melindungi sarangnya. Propolis bersifat desinfektan atau antibakteri sehingga mampu membunuh semua kuman yang masuk ke sarang lebah. Komponen utama dari propolis adalah flavonoid dan asam fenolat, termasuk caffeic acid phenylesthylester (CAPE). Tujuan penelitian ini adalah mengetahui efektivitas ekstrak propolis Trigona sp sebagai antimikroba terhadap bakteri Staphylococcus aureus secara in vitro. Penelitian ini dilakukan menggunakan design eksperimental laboratoris yaitu, metode tube dilution test. Sampel yang digunakan dalam penelitian ini adalah Staphylococcus aureus yang dikultur oleh Laboratorium Mikrobiologi Universitas Brawijaya Malang. Sampel kemudian dibiakkan dan diberi ekstrak propolis Trigona sp dengan konsentrasi 1,4%, 1,2%, 1,0%, 0,8%, 0,6%, 0,4% dan 0,2%. Kemudian diinkubasi selama 24 jam lalu dihitung jumlah pertumbuhan koloni yang ada. Hasil dari penelitian didapatkan Kadar Hambat Minimum (KHM) dari ekstrak propolis Trigona sp adalah 1,0% dan Kadar Bunuh Minimum (KBM) adalah 1,2%. Dari hasil penelitian dapat disimpulkan bahwa ekstrak propolis Trigona sp dapat menghambat pertumbuhan bakteri Staphylococcus aureus secara in vitro.

English Abstract

Staphylococcus aureus found as normal flora in healthy individual oral cavity especially in pulp, but this bacteria can cause an infections if there is disturbance of balance. Attempt in solve the problem of pulp necrosis are root canal treatment and antibiotic using which is inhibit the proliferation of Staphylococcus aureus. Propolis is a substance produced by bees to protect their hive. Propolis have disinfection or antibacterial effect to all the germs that go into this hive. The main components of propolis are flavonoids and phenolic acids, included caffeic acid phenylesthylester (CAPE). The objective of this research was to determine the effectiveness of Trigona sp propolis extract as an antimicrobial against Staphylococcus aureus in vitro. This study was conducted using a laboratory experimental design ie, dilution test tube method. The sample used in this study were Staphylococcus aureus cultured in the Microbiology Laboratory of the Brawijaya University Malang. Trigona sp propolis extract with a concentration of 1,4%, 1,2%, 1,0%, 0,8%, 0,6%, 0,4% and 0,2%. Then incubated in BHI agar for 24 hours and then counted the number of growth existing colonies. The results of the study indicated 1.0% of MIC (Minimum Inhibitory Concentration and 1.2% of MBC (Minimum Bactericidal Concentration) of propolis extract Trigona sp. It can be concluded that Trigona sp propolis extract can inhibit the growth of Staphylococcus aureus in vitro.
