Abstract
Staphylococcus aureus adalah bakteri yang paling sering menginfeksi manusia. Frekuensi infeksi S. aureus nosokomial maupun yang didapat di masyarakat terus meningkat. Anggur merupakan salah satu buah yang dikenal oleh manusia sejak lama dan cukup populer di Indonesia. Anggur Bali merupakan anggur yang paling banyak ditanam dan telah banyak dikenal oleh masyarakat Indonesia. Kulit anggur mengandung senyawa aktif seperti antosianin, tanin, dan saponin yang memiliki aktivitas sebagai antibakteri. Penelitian ini dilakukan untuk mengetahui efektivitas ekstrak kulit anggur Bali (Vitis vinifera L. var. Alphonso lavalle) sebagai antibakteri terhadap S. aureus . Sampel yang digunakan adalah 4 isolat bakteri S.aureus dari pasien yang berbeda. Pembuatan ekstrak kulit anggur Bali dilakukan melalui proses maserasi menggunakan pelarut etanol 96%. Metode penelitian ini menggunakan dilusi agar karena hasil ekstrak berwarna gelap (ungu kehitaman). Variasi konsentrasi ekstrak yang digunakan sebesar 1%; 1,5%; 2%; 2,5%; dan 3% v / v . Pada konsentrasi 2,5% v / v tidak didapatkan pertumbuhan keempat isolat bakteri. Hasil uji statistik non-parametrik Kruskall-Wallis menunjukkan adanya perbedaan efek antibakteri pada setiap perlakuan konsentrasi ekstrak kulit anggur Bali terhadap pertumbuhan koloni S. aureus (p < 0,05). Uji korelasi Spearman menunjukkan semakin tinggi konsentrasi ekstrak kulit anggur Bali, maka semakin rendah pertumbuhan koloni S. aureus (Spearman rho = -0,971; p < 0,05). Kesimpulan penelitian ini adalah ekstrak kulit anggur Bali memiliki efek antibakteri terhadap bakteri S. aureus dengan KHM sebesar 2,5% v / v .
English Abstract
Staphylococcus aureus is a bacteria that most commonly infect humans. Frequency of S. aureus nosocomial and community infections continues to increase. Grape is one of the fruits known for a long time and popular in Indonesia. Bali grape is the most widely planted grapes and has been widely recognized by the people of Indonesia. Grape skin contains active compounds such as anthocyanins, tannins, and saponins which have antibacterial activity. This study was conducted to determine the effectiveness of Bali grape skin extract ( Vitis vinifera L. var. Alphoso lavalle ) as antibacterial against S. aureus . The samples used were 4 isolates of S. aureus from different patients. Preparation of Bali grape skin extracts done through a process of maceration using 96 % ethanol. This research method using a dilution of the extract in order for dark colored (purple-black). Variations in the concentration of extract that is used by 1% , 1,5% , 2% , 2,5% and 3% v / v . At a concentration of 2,5 % v / v was not obtained growth of four bacterial isolates. The results of non-parametric statistical, Kruskall-Wallis test showed the differences in the antibacterial effect of each concentration Bali grape skin extract on the growth of colonies of S. aureus (p < 0.05). Spearman correlation test showed the higher concentrations of Bali grape skin extracts, the lower the growth of colonies of S. aureus (Spearman rho = -0,971, p < 0.05). The conclusion of this study is Bali grape skin extracts have antibacterial effects against S. aureus with a MIC of 2,5 % v / v .
