Abstract
Streptococcus pyogenes merupakan salah satu bakteri penyebab tersering pada inflamasi lanjutan setelah terjadinya pulpitis pada gigi. Untuk mengatasi penyakit infeksi tersebut, banyak dikembangkan penggunaan antibakteri, termasuk antibakteri herbal. Salah satu yang diduga mengandung antibakteri adalah daun alpukat. Daun alpukat telah diketahui mengandung alkaloid, flavonoid dan polifenol yang memiliki efek antibakteri. Penelitian ini bertujuan untuk membuktikan bahwa ekstrak daun alpukat memiliki pengaruh terhadap Streptococcus pyogenes secara in vitro. Penelitian ini merupakan suatu penelitian eksperimental yang dilakukan terhadap Streptococcus pyogenes dengan metode dilusi tabung . Kelompok perlakuan yaitu kelompok bakteri yang diberi ekstrak daun alpukat dengan konsentrasi 0.2%, 0.4%, 0.6%, 0.8%, dan 1%. Kelompok kontrol terdiri dari kontrol bakteri sebagai kontrol positif dan kontrol ekstrak daun alpukat sebagai kontrol negatif. Hasil penelitian menunjukkan bahwa Kadar Hambat Minimal (KHM) tidak dapat diamati, sedangkan Kadar Bunuh Minimal (KBM) diperoleh pada 1%. Analisa data menggunakan One-way ANOVA dengan = 0,05 menunjukkan terdapat perbedaan pengaruh tiap perlakuan konsentrasi ekstrak daun alpukat terhadap jumlah koloni Streptococcus pyogenes . Uji korelasi Pearson menunjukkan semakin tinggi konsentrasi ekstrak daun alpukat, maka semakin rendah pertumbuhan Streptococcus pyogenes secara in vitro . Kesimpulan pada penelitian ini yaitu ekstrak daun alpukat mempunyai pengaruh terhadap Streptococcus pyogenes, dengan KBM 1%.
English Abstract
Streptococcus pyogenes is one of the bacteria that causes exacerbation after the occurrence of pulpitis in a tooth. In order to eliminate the infection, antibacterial agents are developed, including those of herbal origins. One of the herb that suppose to have an antibacterial activity is avocado leaves . Avocado leaves is claimed to contain alkaloid, flavonoid and polifenol that have antibacterial effect. This research is conducted to prove the influence of Avocado leaves extract on Streptococcus pyogenes using in vitro Method . This research is an experimental study which is carried out to Streptococcus pyogenes with tube dilution method. The treated groups are bacteria which are given the avocado leaves extract with range of concentration as follows 0.2%, 0.4%, 0.6%, 0.8%, and 1%. The control groups includes bacteria as positive control and avocado leaves extract as negative control. The result indicated that minimal inhibitory concentration (MIC) can’t be found and minimal bactericidal concentration (MBC) is found at 1%. Results of One-way ANOVA with = 0,05, showed significant difference among each concentration of avocado leaves extract related to the growth of colonies of Streptococcus pyogenes . Pearson correlation test showed that the higher concentration of avocado leaves extract, the less number of colony growth. The conclusion of this study is that avocado leaves extract has influence against Streptococcus pyogenes , with MBC is 1%.
