Abstract
Salmonella Typhi adalah salah satu patogen penyebab penyakit demam tifoid. S.Typhi dilaporkan telah resisten terhadap beberapa obat antibakteri sehingga perlu dilakukan penelitian untuk menemukan alternatif terapi lain. Salah satu alternatif terapinya adalah menggunakan bahan alami sebagai bahan antibakteri, yaitu daun sirih merah (Piper crocatum). Kandungan aktif yang diduga bermanfaat sebagai antibakteri adalah flavonoid, alkaloid, tanin, dan minyak atsiri. Penelitian ini bertujuan untuk mengetahui potensi antibakteri ektrak etanol daun sirih merah terhadap S.Typhi. Penelitian ini merupakan penelitian eksperimental laboratorik dengan menggunakan dilusi tabung untuk menentukan KHM dan KBM. Sampel yang digunakan dalam penelitian ini adalah isolat S.Typhi yang diperoleh dari Laboratorium Mikrobiologi Fakultas Kedokteran Universitas Brawijaya, Malang, Jawa Timur. Konsentrasi yang digunakan dalam penelitian yaitu 26%, 28%, 30%, 32%, 34%, dan 36% dengan empat kali pengulangan, sedangkan konsentrasi S.Typhi adalah 106 CFU/ml. Hasil uji statistik menunjukan bahwa ekstrak etanol daun sirih merah, secara signifikan dapat menghambat pertumbuhan S.Typhi (Kruskal Wallis, p<0.05) dan terdapat hubungan antara peningkatan konsentrasi ekstrak etanol daun sirih merah dengan penurunan jumlah pertumbuhan koloni S.Typhi (R=-0.992). KHM ekstrak etanol daun sirih merah terhadap S.Typhi adalah 30% dan KBM-nya adalah 36%. Dengan demikian, dapat disimpulkan bahwa ekstrak etanol daun sirih merah memiliki efek antibakteri terhadap S.Typhi secara in vitro.
English Abstract
Salmonella Typhi is one of the pathogen cause typhoid fever. S.Typhi is reported resistant to the several antimicrobial drugs and need to be conducted to find other alternative therapy. One of alternative therapy is the use of natural material of the Piper crocatum leaves. The active substance suspected in the Piper crocatum leaves having antibacterial effect are, flavonoid, alkaloid, tanin, and atsiri oil. The experiment aimed to understanding the antibacterial effect of ethanolic extract piper crocatum leaf againts S.Typhi. The experiment of laboratoric experimental is used tube dilution test to determine the Minimum Inhibitory Concentration (MIC) and Minimum Bactericide Concentration (MBC). The sample used in this experiment was S.Typhi obtained from the laboratory of Microbiology, Faculty of Medicine, Brawijaya University, Malang. East of Java. The extract concentration used were 26%, 28%, 30%, 32%, 34%, and 36% with four repetition, while the concentration of S.Typhi was 106 CFU/ml. The statistic analysis showed that ethanolic extract of Piper crocatum can inhibit the growth of S.Typhi significantly (Kruskal Wallis, p<0.05) and there was a relationship between the increasing of ethanolic extract of Piper crocatum leaves with the decreasimg of S.Typhi colony number (R=-0.992). MIC of ethanolic extract of piper crocatum leaves againts S.Typhi was 30% and MBC was 36%. In conclusion, that the ethanolic extract of Piper crocatum leaves has an antibacterial effect againts S.Typhi in vitro.
