Abstract
Lactobacillus acidophilus merupakan bakteri rongga mulut yang menghasilkan zat asam yang dapat menimbulkan karies gigi. Karies gigi merupakan salah satu keadaan patologis dari gigi. Salah satu penggunaan rimpang kencur sebagai bahan baku alternatif pencegahan karies gigi. Rimpang kencur (Kaempferia galanga L.) mengandung zat antibakteri berupa sineol, borneol, dan kamfer. Tujuan penelitian ini adalah untuk membuktikan bahwa ekstrak metanol rimpang kencur memiliki efek antibakteri terhadap Lactobacillus acidophilus secara in vitro. Penelitian ini merupakan eksperimental laboratorik dengan metode dilusi tabung untuk mendapatkan nilai KHM dan KBM. Konsentrasi ekstrak metanol rimpang kencur yang digunakan adalah 17,5%, 18,75%, 20%, 21,25%, dan 22,5%. Penelitian ini menggunakan 5 konsentrasi dengan pengulangan masing-masing konsentrasi 4 kali. Nilai KHM diperoleh dengan cara mengamati perbedaan tingkat kekeruhan tiap konsentrasi larutan ekstrak metanol rimpang kencur. Nilai KBM dapat diperoleh dengan mengamati pertumbuhan bakteri tiap konsentrasi pada media agar. Hasil penelitian menunjukkan Kadar Hambat Minimum (KHM) diperoleh pada konsentrasi 21,25%, sedangkan Kadar Bunuh Minimum (KBM) pada konsentrasi 22,5%. Analisis data menggunakan oneway ANOVA menunjukkan adanya perbedaan yang signifikan pada perubahan konsentrasi ekstrak metanol rimpang kencur terhadap jumlah koloni Lactobacillus acidophilus (p<0,01). Uji korelasi Pearson menunjukkan adanya hubungan yang kuat dan berbanding terbalik antara konsentrasi ekstrak metanol rimpang kencur dengan jumlah koloni Lactobacillus acidophilus (-0,934). Uji regresi menunjukkan efektifitas ekstrak metanol rimpang kencur terhadap Lactobacillus acidophilus sebesar 87,3% (R square = 0,873). Berdasarkan penelitian ini, dapat disimpulkan bahwa ekstrak metanol rimpang kencur mempunyai efek antibakteri terhadap Lactobacillus acidophilus secara in vitro.
English Abstract
Lactobacillus acidophilus is an oral bacterium that produces acid causing dental caries. Dental caries is one pathological condition that affects the tooth. One use greater galingale rhizome as alternative raw materials prevention of dental caries.. The greater galingale (Kaempferia galanga L.) rhizome contain antibacterial substances such as sineol, borneol, and chamfer. The purpose of this study is to prove that the methanol extract of greater galingale rhizome have antibacterial effects against Lactobacillus acidophilus in vitro. This is a laboratory experimental study using tube dilution method to find MIC and MBC. The used concentration of methanol extract of greater galingale rhizome are 17,5%, 18,75%, 20%, 21,25%, and 22,5%. This experiment use 5 concentration with 4 repetition for each concentration. MIC values obtained by observing the difference in the level of turbidity of each solution concentration of methanol extract of greater galingale rhizome. MBC value can be obtained by observing the growth of bacteria on an agar medium for each concentration. The result shows the Minimum Inhibitory Concentration (MIC) is concentration of 21,25%, while the Minimum Bactericidal Concentration (MBC) is concentration of 22,5%. Analyzing oneway ANOVA shows a significant difference in the change of concentration of methanol extract of greater galingale rhizome on the number of colonies of Lactobacillus acidophilus (p <0.01). Pearson correlation test shows a strong and inversely relationship between the concentration of methanol extract of greater galingale rhizome and the number of colonies of Lactobacillus acidophilus (-0,934). Regresion test show efectivity methanol extract of greater galingale rhizome againts Lactobacillus acidophilus is 87,3% (R square = 0.873). The conclusion from this experiment is methanol extract of greater galingale have antibacterial effects againts Lactobacillus acidophilus in vitro.
