Abstract

Ulkus peptikum adalah suatu daerah ekskoriasi mukosa lambung atau usus, yang dapat timbul sebagai efek samping penggunaan NSAIDs. Penghambatan COX-1 secara sistemik oleh NSAIDs dapat menurunkan faktor perlindungan lambung. Jintan hitam (Nigella sativa L.) mengandung zat aktif yakni thymoquinone (TQ) yang dapat membantu memperbaiki pertahanan mukosa lambung dan membantu penyembuhan ulkus peptikum. Tujuan penelitian ini adalah untuk membuktikan efek pemberian ekstrak jintan hitam (Nigella sativa L.) dalam mengurangi tingkat kerusakan mukosa lambung pada tikus akibat induksi Indometasin. Penelitian ini merupakan eksperimental murni dengan metode The Posttest Control Group Design. Sample terdiri dari 25 ekor tikus Rattus novergicus strain Wistar jantan, dipilih dengan simple random sampling menjadi lima kelompok. Masing-masing kelompok dipuasakan selama 24 jam. Kelompok 1 (negatif) mendapat aquabides secara oral, kelompok 2 (positif) mendapat Indometasin 30 mg/kgBB, kelompok 3 mendapat Indometasin 30 mg/kgBB dan ekstrak jintan hitam 100 mg/kgBB, kelompok 4 mendapat Indometasin 30 mg/kgBB dan ekstrak jintan hitam 200 mg/kgBB, dan 5 mendapat Indometasin 30 mg/kgBB dan ekstrak jintan hitam 300 mg/kgBB. Ekstrak jintan hitam diberikan setiap 8 jam selama 24 jam. Gaster tikus diamati dan diklasifikasikan berdasarkan skor 8 jam setelah perlakuan. Data yang diperoleh dianalisis dengan uji statistik One-way ANOVA dilanjutkan Post Hoc LSD dengan α=0,05. Hasil analisis One-way ANOVA menunjukkan perbedaan yang signifikan pada rata-rata ulkus peptikum lambung (p<0,05). Hasil Post Hoc LSD menunjukkan perbedaan yang signifikan antara kontrol positif dengan kelompok perlakuan. Kesimpulan dari penelitian ini adalah pemberian ekstrak jintan hitam mampu mengurangi tingkat kerusakan mukosa lambung pada tikus

English Abstract

Peptic ulcer is an area of the stomach or intestinal mucosal excoriation, which may arise as a side effect of NSAIDs use. Inhibition of COX-1 systemically by NSAIDs may reduce gastric protection factor. Black cumin (Nigella sativa L.) containing the active substance thymoquinone (TQ) which can help improve gastric mucosal defense and help healing of peptic ulcer. The purpose of this study was to prove the effects of extract of black cumin (Nigella sativa L.) in reducing gastric mucosal damage levels of the Indomethacin-induced rats. This study is a purely experimental method The posttest control group design. Sample consisted of 25 rats Rattus novergicus male Wistar strain, selected by simple random sampling into five groups. Each group was fasted for 24 hours. The 1st group (negative) received orally aquabides, 2nd group (positive) received Indomethacin 30 mg/kgBB, 3rd group received Indomethacin 30 mg/kgBB and black cumin extract 100 mg/kgBB, 4th group received Indomethacin 30 mg/kgBB and black cumin extract 200 mg/kgBB, and 5th group received Indomethacin 30 mg/kgBB and black cumin extract 300 mg/kgBB. Extracts of black cumin was given every 8 hours for 24 hours. Rat gastric observed and classified by a score 8 hours after treatment. Data were analyzed with statistical tests One-way ANOVA followed by Post Hoc LSD α = 0.05. Results of One-way ANOVA analysis showed a significant difference in the average gastric peptic ulcers (p <0.05). Results of Post Hoc LSD showed a significant difference between the positive control group with treatment groups. The conclusion of this experiment is that giving black cumin extracts is able to reduce the level of gastric mucosal damage of the rats
