Efek Pemberian Ekstrak Jintan Hitam (Nigella sativa L.) Terhadap Kedalaman Lesi Mukosa Lambung yang Diamati Secara Mikroskopis pada Tikus Rattus novergicus Strain Wistar yang Diinduksi Indometasin

Abstract

Lambung memiliki sawar mukosa yang berfungsi untuk melindungi dari cedera mekanis, cedera karena zat asam, dan untuk mencegah lambung mencerna dirinya sendiri. Ketidakseimbangan antara faktor agresif dan defensif akibat penggunaan obat golongan Non Steroid Antiinflamatory Drugs (NSAIDs) non selektif, seperti indometasin, akan menyebabkan kerusakan pada mukosa lambung. Manfaat ekstrak jintan hitam (Nigella sativa L.) yang mengandung zat aktif Thymoquinone dipercaya memiliki efek antiinflamasi, antioksidan, gastroprotektif, dan lain-lain. Untuk membuktikannya, dilakukan studi eksperimental menggunakan the post test control group design kepada 25 ekor tikus Rattus novergicus strain wistar jantan untuk mengetahui hasil terhadap pengurangan kedalaman lesi mukosa lambung. Simple random sampling dilakukan untuk membagi tikus ke 5 kelompok, yaitu kontrol positif, kontrol negatif, perlakuan dosis 1, perlakuan dosis 2, dan perlakuan dosis 3. Hasil penelitian ini menunjukkan data terdistribusi normal melalui Uji Kolmogorov-Smirnov Test dengan nilai p>0,05. Terdapat perbedaan yang signifikan terhadap skor integritas sel epitel mukosa lambung dengan nilai p=0.000 pada uji ANOVA (signifikan jika p<0,05). Hasil Pos Hoc Tukey menunjukkan bahwa ada perbedaan signifikan pada skor integritas epitel mukosa antar 2 kelompok, kecuali antara kontrol positif dengan perlakuan dosis 1 dan kontrol negatif dengan perlakuan dosis 3. Uji Korelasi Pearson menunjukan nilai r=-0,953, yang berarti data korelasi negatif yang sangat kuat. Artinya semakin tinggi dosis ekstrak jintan hitam yang diberikan, maka semakin sedikit kerusakan sel epitel mukosa lambung. Hasil uji regresi linier menunjukkan pengaruh variabel independen (ekstrak jintan hitam) lebih berperan 90,4% terhadap variabel dependen (skor integritas epitel lambung), dibandingkan oleh pengaruh faktor eksternal. Terbukti bahwa ekstrak jintan hitam dapat mengurangi kedalaman lesi mukosa lambung.

English Abstract

Gastric mucosal barrier function has to protect from mechanical injury, injury due to acid substances, and to prevent the stomach digest itself. The imbalance between aggressive and defensive factors because the use of medication Non Steroid Antiinflamatory Drugs (NSAIDs) non-selective, such as indomethacin, will cause damage to the gastric mucosa. Benefits of black cumin (Nigella sativa L.) extracts that containing the active substance, Thymoquinone, believed to have anti-inflammatory, antioxidant, gastroprotective, and others functions. To prove it, we conducted an experimental study using the post-test control group design to 25 male rats of Rattus novergicus strain wistar to know the result of the reduction of gastric mucosal lesion depth. Simple random sampling is to divide the rats into 5 groups: positive control, negative control, treatment dose 1, treatment dose 2, and treatment dose of 3. The results showed normally distributed data via the Kolmogorov-Smirnov test with p> 0,05. There are significant differences on scores of gastric mucosal epithelial cell integrity with p = 0.000 on ANOVA test (significant if p <0.05). Results of Post Hoc Tukey showed that there were significant differences in mucosal epithelial integrity scores between the 2 groups, except for the positive control with treatment doses of 1, and a negative control with treatment dose 3. The Pearson shows value of correlation test r = -0.953, which means that a negative correlation data very strong. This means that the higher dose of the extract of black cumin are given, the less damage to the gastric mucosal epithelial cells. Results of linear regression shows the effect of the independent variable (black cumin extract) more involved 90.4% of the dependent variable (score of gastric epithelial integrity), compared to the influence of external factors. We can conclude that give black cumin extract can reduce gaster mucosal lesion depth.
