Abstract

Pengobatan yang efektif di daerah genitalia wanita harus memiliki sistem penghantaran obat yang sesuai dengan kondisi daerah vagina yang memiliki aktivitas self-cleansing. Penggunaan kombinasi polimer mukoadhesive dan superdisintegaran Crosscarmellose Sodium diharapkan dapat membantu penghantaran obat yang sesuai dengan kondisi vagina. Tujuan penelitian ini adalah untuk mengetahui kadar kombinasi polimer HPMC dan superdisintegran Crosscarmellose Sodium untuk menghasilkan daya adhesi dan daya pelepasan obat dalam tablet vaginal Metronidazole yang optimum. Metode pembuatan tablet yang digunakan yakni metode kempa langsung. Pada penelitian menggunakan 3 formula yang mempunyai kadar kombinasi yang berbeda yakni dengan perbandingan kadar kombinasi untuk formula 1 sebesar 30:1, formula 2 sebesar 30:3 dan formula 3 sebesar 50:1. Dari hasil uji yang telah dilakukan, menunjukkan bahwa untuk uji IPC dan karakteristik fisik tablet seperti laju alir, homogenitas, keseragaman bobot, keseragaman ukuran, kekerasan, kerapuhan, penetapan kadar dan waktu hancur memenuhi persyaratan penerimaan tablet. Sedangkan uji laju pelepasan obat dengan kadar formula 1 memberikan pelepasan obat sebesar 0,1257 mg/menit setelah 4 jam jika dibandingkan dengan formula 2 dan formula 3. Daya adhesive yang paling tinggi terdapat pada F3 dengan konsentrasi HPMC sebesar 50%. Akan tetapi ketiga formula masih memiliki daya adhesive yang masih diterima oleh persyaratan (minimal 3 jam di area vagina). Berdasarkan hasil tersebut kadar kombinasi formula yang optimum terdapat pada formula 1 yang mempunyai laju pelepasan yang cepat dan daya adhesive yang mampu menghambat pelepasan dalam waktu 4 jam.

English Abstract

Effective treatment of female genitalia should have a drug delivery system in according to the conditions of the vagina has a self-cleansing activity. The use of a combination of polymer mukoadhesive HPMC and superdisintegaran Crosscarmellose Sodium expected to help drug delivery in according to the conditions of the vagina. The purpose of this study was to determine the levels optimum combinations of HPMC and Sodium Crosscarmellose against power adhesion and drug release in Metronidazole vaginal tablets. Method for making a tablet that used the clamp method directly. In studies using 3 formula with different levels of combination, formula 1 consist of 30: 1, formula 2 consist of 30: 3 and formula 3 consist of 50: 1. From the test results that have been conducted, showing for IPC test and physical characteristics of tablets such as flow rate, homogeneity, uniformity of weight, size uniformity, hardness, friability, disintegration time assay and meet the admission requirements of tablets. While the rate of drug release test, formula 1 provide drug release at 0.1257 mg/min after 4 hours when compared with formula 2 and formula 3 adhesive highest power contained in F3 with HPMC concentration by 50%. However formula 1 still have the adhesive power because it is accepted by the requirements (a minimum of 3 hours in the vaginal area). Based on these results the optimum levels of the combination formula contained in the formula 1 which has a fast release rate and power adhesive is capable of inhibiting the release within 4 hours.
