Abstract

Tingginya prevalensi infeksi cacing Ascaris lumbricoides di Indonesia membutuhkan upaya penanggulangan yang intensif karena mengakibatkan komplikasi yang cukup berat. Pada era ini, masyarakat lebih memilih bahan ramuan dan obat-obatan alami untuk pengobatan. Salah satu tanaman yang berpotensi menjadi obat alternatif adalah tumbuhan Sambiloto (Andrographis paniculata). Ekstrak daun sambiloto diketahui memiliki kandungan zat aktif seperti tanin, saponin dan andrografolid yang terbukti memiliki daya anthelmintik. Penelitian ini bertujuan untuk mengetahui daya anthelmintik ekstrak metanol daun sambiloto (Andrographis paniculata) terhadap Ascaris suum secara in vitro. Penelitian ini menggunakan Ascaris suum, karena secara etis lebih mudah didapat dan memiliki genus, kemiripan morfologi, cara infeksi dan patomekanisme yang sama dengan Ascaris lumbricoides. Penelitian ini bersifat eksperimental laboratorium dengan rancangan penelitian post test only controlled group design. Sampel terbagi dalam lima kelompok perlakuan, yaitu kontrol negatif yang diinkubasi dalam larutan NaCl 0.9%, kontrol positif dengan larutan Pirantel Pamoat 1%, dan kelompok perlakuan dalam larutan ekstrak daun sambiloto dengan tiga konsentrasi, yaitu 20%, 40%, dan 60%. Cacing diamati dalam 11 waktu pengamatan, yaitu jam pertama, ke-2, 3, 4, 5, 6, 7, 8, 9, 10, dan jam ke 24, dan selanjutnya data dianalisis statistik dengan uji analisis statistik Regresi Probit menggunakan program IBM SPSS for Windows, Version 22.0. Dari hasil uji Regresi Probit didapatkan LC100 ekstrak daun sambiloto pada konsentrasi ekstrak 38,5% dan LT100 ekstrak metanol daun sambiloto konsentrasi 40% adalah pada waktu 9 jam dan 40 menit. Kesimpulan penelitian ini adalah menunjukkan bahwa ekstrak daun sambiloto (Andrographis paniculata) memiliki efek anthelmintik terhadap Ascaris suum secara in vitro.

English Abstract

The prevalence of Ascaris lumbricoides infection in Indonesia is still high. Disease prevention efforts must be intensified because of possible severe complication. Sambiloto leaf is expected to be an alternative medicine, as it has active substances such as tannin, saponnin and andrografolid that are proven to have efficacy as anthelmintic. The aim of this study is to observe the efficacy of Sambiloto leaf (Andrographis paniculata) methanol extract as anthelmintic againts Ascaris suum in vitro. This study used Ascaris suum instead of Ascaris lumbricoides, because of their high similarity. Besides, Ascaris suum is much easier to get. This study was an experimental laboratory study with post test only controlled group design. The samples were divided into five groups: negative control group (NaCl 0.9 %), positive control group (pyrantel pamoat solution 1%), and experimental groups with several concentration of sambiloto leaf extract (20%, 40%, and 60%). The worms were observed for 11 hours; started at 1st hours, 2nd, 3rd ,4th, 5th, 6th, 7th, 8th, 9th,10th, and finally 24th hours. Obtained data was analyzed by Probit Regression analysis using IBM SPSS 22.0. We found that lethal concentration 100 (LC100) of sambiloto leaf methanol extract was at concentration 38,5%. We found that Lethal Time 100 (LT100) of 40% extract sambiloto leaf was at 9 hours 40 minutes. This study proved that sambiloto leaf methanol extract (Andrographis paniculata) has anthelmintic effect againts Ascaris suum in vitro.
