Abstract

Daun Kelor (Moringa oleifera) memiliki kandungan utama senyawa Flavonoid yaitu quercetin dan kaempherol yang memiliki efek antikanker. Sel MCF-7. Pada kanker payudara terdapat ekspresi yang berlebihan dari Hsp70 yang berperan dalam mekanisme perlindungan sel dari berbagai kondisi fisiologis yang mengakibatkan kerusakan sel. Sehingga meningkatkan pertumbuhan sel kanker dan menghambat terjadinya apoptosis. Penelitian ini bertujuan membuktikan bahwa ekstrak daun kelor dapat menurunkan ekspresi Hsp70 dan meningkatkan apoptosis sel MCF-7. Penelitian ini dilakukan dengan True experimental in vitro design pada sel kanker MCF-7. Penentuan IC50 dilakukan dengan metode MTT Assay dan dipaparkan ekstrak dengan rentang konsentrasi 1200 – 20000 μg/ml. Uji ekspresi Hsp70 dan apoptosis menggunakan kelompok perlakuan yaitu, kontrol negatif, perlakuan konsentrasi ekstrak 1100, 2200, 4400 μg/ml. Hasil IC50 yang diperoleh yaitu sebesar 2200 μg/ml. Terdapat perbedaan signifikan pada perlakuan konsentrasi, 1100 μg/ml yaitu 71,22% (p=0,000), 2200 μg/ml yaitu 38,38% (p=0,000), dan 4400 μg/ml yaitu 64,39 % p=0,000). Hasil uji apoptosis tidak dapat analisis karena sel yang berhasil diamati sangat sedikit. Namun, diamati dari degradasi warna, menunjukkan apoptosis cenderung mengalami peningkatan. Kesimpulan dari penelitian ini adalah IC50 ekstrak daun kelor sebesar 2200 μg/ml. Ekstrak daun kelor dapat menurunkan ekspresi Hsp70 dan apoptosis cenderung meningkatan seiring dengan peningkatan konsentrasi ekstrak.

English Abstract

Moringa oleifera leaves contain the predominant flavonoids. There are quercetin and kaempferol that have anticancer effect. Breast cancer cells are expressed high levels of Hsp70 in response to protect cells from variety of physiological lead to lethal conditions. It cause cell proliferation and inhibition of apoptosis. The aims of this study were to show Moringa oleifera leaves extract effect on Hsp70 expressions, apoptosis of MCF-7 cells and determine IC50 of Moringa oleifera leaves extract. IC50 was determined by MTT Assay using Moringa oleifera leaves extract concentration range 1200–20000 μg/ml. Hsp70 expression and apoptosis assay were performed to control, 1100, 2200, and 4400 μg/ml extract group. Results showed that Hsp70 expressions decreased significantly in the group of 1100 μg/ml for 71.22% (p=0.000), 2200 μg/ml for 38.38% (p=0.000), and 4400 μg/ml for 64.39% (p=0.000). The results of the apoptotis assay could not be analyzed because the number of apoptotic cells could be observed were very little. However, they showed inclination increase by observed of the color gradation of the cells. The conclusions of this study were Moringa oleifera leaves extract IC50 was 2200 μg/ml. Moringa oleifera leaves extract decreased Hsp70 expressions and apoptosis cells showed inclination increase by increasing the extract concretation.
