Abstract

Aterosklerosis merupakan suatu kondisi yang merupakan suatu respons keradangan kronik terhadap deposisi lipoprotein pada dinding arteri. Diet aterogenik memicu terjadinya respon keradangan yang ditandai oleh peningkatan aktivasi Nuclear Factor Kappa B (NF-kB). NF-kB merupakan faktor transkripsi yang memicu inflamasi pada pembuluh darah (endotel). Lectin Like LDL Receptor 1 (LOX-1) merupakan reseptor Ox-LDL pada sel endotel yang berperan penting dalam patogenesis aterosklerosis. Penelitian ini bertujuan menentukan pengaruh pemberian protein LOX-1 terhadap aktivasi NF-kB tikus Wistar yang diberi diet aterogenik. Penelitian ini merupakan penelitian eksperimental laboratorik menggunakan postest only control group design yang dilakukan 56 hari terhadap 28 tikus Wistar jantan. Tikus dibagi dalam 7 kelompok penelitian yaitu kontrol negatif yang diberi diet normal, kontrol positif yang diberi diet aterogenik, kelompok perlakuan yang diberi aterogenik dan protein LOX-1 dengan dosis 1 ng+alum (P1), 10 ng+alum (P2), 100 ng+alum (P3), 1 μg+alum (P4), dan alum saja (P5). Pada akhir peneitian, tikus diambil jaringan aorta untuk menentukan aktivasi NF-kB dengan metode immunohistokimia. Analisis dengan uji One-Way ANOVA menunjukkan penurunan aktivasi NF-kB (p = 0,000). Terdapat korelasi negatif kuat antara pemberian protein LOX-1 dengan aktivasi NF-kB (p = 0,000, r = - 0,837, Uji Pearson). Kesimpulan dari penelitian ini adalah pemberian protein LOX-1 pada dosis 1 μg menurunkan aktivasi NF-kB.

English Abstract

Atherosclerosis is a condition chronic inflammatory condition as a response to the lipoproteins deposition in the artery wall. Atherogenic diet triggers inflammatory response characterized by Nuclear Factor Kappa B (NF-kB) activation increasing. NF-kB is a transcription factor that triggers inflammation in endothelium. Lectin Like LDL Receptor 1 (LOX-1) is a scavanger receptor of Ox-LDL on endothelial cells that play an important role in the pathogenesis of atherosclerosis. The aim of this studyis to determine the effect of admission of LOX-1 protein to NF-kB activation Wistar rats fed atherogenic diet. This study was experimental laboratory with postest only control group design for 56 days using 28 male Wistar rats. Rats were divided into 7 groups which were negative control with normal diet, positive control with atherogenic diet, atherogenic diet and LOX-1 protein treatment with dosage 1 ng + alum (P1). 10 ng + alum (P2), 100 ng + alum (P3), 1μg + alum (P4) and alum only (P5). At the end of the study, aorta tissues were taken to determine its NF-kB activation with immunohistochemical method. One-Way ANOVA results test showed that there NF-kB activation decresed (p = 0.000). Furthermore, there was a strong negative correlation between the admission of LOX-1 and NF-kB activation (p = 0.000, r = - 0.837, Pearson test). The conclusion of this study was that the admission LOX-1 protein with dosage 1 μg was decreases NF-kB activation.
