Abstract

Aktivasi NF- ĸB (parameter apoptosis kanker) menyebabkan induksi beberapa fungsi seluler antara lain proliferasi sel meningkat dan penurunan apoptosis. Daun kelor (Moringa oleifera) secara ayurveda telah terbukti dalam mencegah penyakit leukemia dan skin papilomagenesis. Tujuan penelitian ialah membuktikan bahwa pemberian ekstrak daun kelor menurunkan aktivitas sel kanker payudara MCF-7. Penelitian menggunakan True experimental in vitro design dengan sel MCF-7. MTT Assay dilakukan dengan pemberian terapi dengan dosis dari rentang 7,1825 – 20000 μg/ml untuk menentukan IC50 pada sel MCF-7. IC50 yang didapatkan adalah dosis 2200 μg/ml. Immunositokimia dan TUNEL assay dengan kelompok kontrol negatif, kelompok perlakuan dosis 1100, 2200, 4400 μg/ml. Terdapat perbedaan signifikan yang lebih rendah pada perlakuan dosis 2200 μg/ml (p=0,000) dan dosis 4400 μg/ml (p=0,000) dibanding kontrol tanpa terapi pada aktivitas NF ĸB. Besar persentase aktivitas NF ĸB pada dosis 2200 μg/ml =31,651% dan 4400 μg/ml =36,575%. Tidak dapat dilihat apoptosis dari MCF-7. Kesimpulan dari penelitian ialah ekstrak daun kelor dapat menurunkan aktivitas sel MCF-7, ekstrak daun kelor dapat menurunkan jumlah NF ĸB aktif pada dosis 2200 dan 4400 μg/ml, dosis IC50 yang diperoleh 2200 μg/ml, apoptosis tidak dapat diamati pada penelitian ini.

English Abstract

Activation of NF ĸB (cancer apoptosis parameter) caused induction of some celluler function, including increased of cell proliferation and decreased apoptosis. Moringa leaves (Moringa oleifera) based on ayurvedic reported can prevent leukimia and skin papilomagenesis. The aim of this research is to prove Moringa leaves extract lowers the activity of the breast cancer cells MCF-7. This research is using True experimental in vitro design with cell MCF-7. MTT Assay performed by administering the therapeutic dose range of 7.1825 – 20000 ug / ml extract moringa leaves to determine the IC50 in MCF-7 cells. IC50 obtained was 2200 ug/ml. Immunocytochemystry and TUNEL assay with negative control group, treated with different dosage group 1100, 2200, 4400 ug/ml. There were lower significant differences in therapeutic dose 2200 ug/ml (p=0.000) and dose 4400 ug/ml (p=0.000) compared to controls without therapy on the activity of NF ĸB. The percentage of NF ĸB activity at doses 2200 μg/ml =31.651% and 4400 μg/ml =36.575%. Apoptosis of cells MCF-7 not visible to observed. In conclusions, extract moringa leaves can inhibit activity of MCF-7, IC50 was 2200 μg/ml and extract moringa leaves be able to decreased NF ĸB activity at dosage 2200 and 4400 μg/ml, and apoptosis was not able to observed in this study.
