Abstract
Hiperglikemia kronik pada diabetes melitus mengakibatkan timbulnya kondisi stres oksidatif dimana terjadi ketidakseimbangan antara produksi reactive oxygen species (ROS) dengan mekanisme pertahanan antioksidan tubuh. Jaringan tubuh tersusun dari lipid yang rentan terhadap ROS, akibatnya terjadi peroksidasi lipid terutama pada jaringan jantung yang memiliki regenerasi enzim antioksidan yang lambat sehingga menyebabkan hipertrofi miosit, apoptosis, dan fibrosis intersisial yang berkontribusi dalam penurunan fungsi jantung. Salah satu produk hasil peroksidasi lipid adalah malondialdehyde (MDA). Ekstrak biji jintan hitam mengandung zat aktif thymoquinone yang menunjukkan aktivitas yang mirip dengan superoxide dismutase (SOD) sehingga dapat berperan sebagai antioksidan. Penelitian ini bertujuan untuk menganalisa efek pemberian ekstrak biji jintan hitam terhadap perbedaan kadar MDA jantung pada tikus Wistar model DM tipe 2. Desain penelitian yang digunakan adalah true experimental menggunakan post test-only controlled design. Dua puluh lima ekor tikus dibagi secara simple random sampling dalam lima kelompok, terdiri dari kelompok tikus diabetes dengan metformin 75 mg/kgBB/hari (Pp), tikus diabetes dengan 10% tween 80 sebagai pelarut (Pn), tikus diabetes yang diberi 3 dosis ekstrak biji jintan hitam 24 (P1), 48 (P2), dan 96 (P3) mg/kgBB/hari selama 4 minggu. Untuk mengetahui kadar MDA jantung digunakan metode thiobarbituric acid (TBA). Hasil penelitian menunjukkan bahwa terdapat perbedaan kadar MDA jantung yang lebih rendah secara bermakna pada kelompok P1 (p = 0,004), P2 (p = 0,031) dan Pn (p = 0,011) dibandingkan kelompok Pp. Kesimpulan dari penelitian ini, pemberian ekstrak biji jintan hitam dapat menurunkan kadar MDA jantung tikus model DM tipe 2 secara bermakna dengan dosis optimal 24 mg/kgBB/hari.
English Abstract
Chronic hyperglycemia on diabetes mellitus cause oxidative stress condition that resulted in imbalance production between reactive oxygen species (ROS) and antioxidant defense mechanism in body. Body tissues composed of lipid that is susceptible to ROS, therefore lipid peroxidation especially on heart tissues which has slow antioxidant enzyme regeneration, finally it can end up in myosit hypertrophy, apoptosis, and intertitial fibrosis that contribute to heart function deterioration. One of the lipid peroxidation product is malondialdehyde (MDA). Black seed extract contain thymoquinone which has similiar activity as superoxide dismutase (SOD) so that it can acts as antioxidant. The aim of this research is to analyze the effect of black seed extract toward the difference of heart MDA level of type 2 diabetic model Wistar strain rats. This was a true experimental research with post test only controlled design. Twenty five Wistar rats were divided into five groups: group of diabetic rats with metformin 75 mg/kgBW/day, group of diabetic rats with 10% tween 80 as diluent, group of diabetic rats treated daily with different dosages of black seed extract 24 (P1), 48 (P2), and 96 (P3) mg/kgBW/day for 4 weeks. MDA level of heart tissues were measured by thiobarbituric acid (TBA) method. The research data showed that there were low significant differences between MDA level of heart tissues of P1 (p = 0,004), P2 (p = 0,031), and Pn (p = 0,011) group compared with Pp group. The conclusion of this research was black seed extract could decrease MDA level of heart tissues in type 2 diabetic rats model significantly with the optimum dose 24 mg/kgBW/day.
