Abstract

Aterosklerosis adalah penebalan dinding arteri karena respon inflamasi kronis terhadap cedera endotel yang melibatkan faktor peradangan IL-6. Biji pare (Momordica charantia) mengandung saponin yang terbukti mampu menurunkan kadar IL-6 pada proses aterosklerosis. Penelitian ini bertujuan untuk membuktikan bahwa ekstrak biji pare yang mengandung saponin dapat menghambat peningkatan kadar IL-6 serum. Studi true experimental dengan metode randomized posttest only controlled group design. Total 25 ekor tikus Wistar jantan dibagi menjadi 5 kelompok: kelompok diet aterogenik sebagai kontrol(+), kelompok diet aterogenik dengan ekstrak biji pare dosis 150 μg/gBB, kelompok diet aterogenik dengan esktrak biji pare dosis 300 μg/gBB, kelompok diet aterogenik dengan ekstrak biji pare dosis 500 μg/gBB, serta kelompok diet normal sebagai kontrol(-), masing-masing selama 12 minggu. Ekstrak biji pare diberikan mulai minggu 8. Setelah 12 minggu dilakukan ELISA dengan sampel serum. Selanjutnya dilakukan pengukuran kadar IL-6 serum dengan metode ELISA. Uji One-way ANOVA menunjukkan ada perbedaan bermakna kadar IL-6 serum antar kelompok (p=0,000), sementara uji Tukey menunjukkan bahwa kadar IL-6 kelompok dosis 300 μg/gBB dan kelompok dosis 500 μg/gBB lebih rendah signifikan dibanding kelompok diet aterogenik (p<0,05), dan tidak memiliki perbedaan dibanding kontrol(-) (p>0,05). Hal ini menunjukkan bahwa dosis 300 dan 500 μg/gBB potensial dalam menghambat peningkatan kadar IL-6 serum. Uji korelasi antar kelompok perlakuan diperoleh nilai R=-0,753 (p=0,000). Kesimpulan penelitian ini adalah pemberian ekstrak biji pare (Momordica charantia) terbukti dapat menghambat peningkatan kadar IL-6 serum tikus Wistar dengan diet aterogenik.

English Abstract

Atherosclerosis is artery wall thickening due to chronic inflammation to endothelial injury which involves inflammatory factor IL-6. Meanwhile, bitter melon (Momordica charantia) seed contains saponin, which had been proven reducing IL-6 in atherosclerosis process. This study is aimed to prove that bitter melon seed extract which is thought to contain saponin can inhibit serum IL-6 level. This true experimental study uses randomized posttest only controlled group design. Total 25 rats were devided in 5 groups: atherogenic diet group as control (+), atherogenic diet group with bitter melon seed extract dose of 150 μg/gBB, atherogenic diet group with the seeds of bitter melon extracts dose of μg/gBB, ath-erogenic diet group with bitter melon seed extract dose of 500 μg/gBB, as well as group normal diet as control (-), respectively for 12 weeks. Bitter melon seed extract given from week 8. After 12 weeks, ELISA performed with serum sample. Next step, serum IL-6 level was measured by ELISA method. One-way ANOVA test showed that there was significant difference serum IL-6 level between all groups (p=0,000), while Tukey test showed that IL-6 level in 300 μg/gBB doses group and 500 μg/gBB doses group significantly lower than the atherogenic diet group (p <0.05), and no differences compared to normal diet group (p> 0.05). This suggests that the dose of 300 and 500 μg/gBB potent in inhibiting the elevated levels of serum IL-6. Correlation test between treatment groups showed the value of R=-0,753 (p=0,000). Conclusion of this study is bitter melon (Momordica charantia) seed extract administration was proven inhibiting serum IL-6 level in Wistar rat with atherogenic diet.
