Abstract
Tingginya prevalensi infeksi cacing Ascaris lumbricoides di Indonesia membutuhkan upaya penanggulangan yang intensif karena mengakibatkan risiko infeksi yang cukup berat. Pada era ini, masyarakat lebih memilih bahan ramuan dan obat-obat an alami untuk pengobatan. Salah satu tanaman yang berpotensi menjadi obat alternatif adalah tanaman mimba (Azadirachta indica). Ekstrak daun mimba diketahui, memiliki kandungan zat aktif seperti saponin, flavonoid dan tanin, yang terbukti memiliki daya anthelmintik. Penelitian ini bertujuan untuk mengetahui daya anthelmintik ekstrak etanol daun mimba (Azadirachta indica) terhadap Ascaris suum secara in vitro. Penelitian ini menggunakan Ascaris suum, karena secara etis lebih mudah didapat dan memiliki genus, kemiripan morfologi, cara infeksi dan patomekanisme yang sama dengan Ascaris lumbricoides. Penelitian ini bersifat eksperimental laboratorium dengan rancangan penelitian the post only controlled group design. Sampel terbagi dalam lima kelompok perlakuan, yaitu kontrol negatif yang diinkubasi dalam larutan NaCl 0.9%, kontrol positif dengan larutan Pirantel Pamoat 1%, dan kelompok perlakuan dalam larutan ekstrak daun mimba dengan tiga konsentrasi, yaitu 20%, 30%, dan 40%. Cacing diamati dalam 12 waktu pengamatan, yaitu jam pertama, ke-2, 3, 4, 5, 6, 7, 8, 9, 10, 11 sampai dengan jam ke 24, dan selanjutnya data dianalisis statistik dengan uji analisis statistik Regresi Probit menggunakan program IBM SPSS for Windows, Version 22.0. Dari hasil uji Regresi Probit didapatkan LC100 ekstrak daun mimba pada konsentrasi ekstrak 34,7%. Dan LT100 ekstrak etanol daun mimba konsentrasi 40% adalah pada waktu 15 jam dan 9 menit. Kesimpulan penelitian ini adalah menunjukkan bahwa ekstrak daun mimba (Azadirachta indica) memiliki daya anthelmintik terhadap Ascaris suum secara in vitro.
English Abstract
The prevalence of Ascaris lumbricoides infection in Indonesia is still high. Disease prevention efforts must be intensified because have the severe risk. Mimba leaf is expected to be one of the alternative medicine, as it has active substances such as saponnin, flavonoids and tannins that are believed to have the efficacy as anthelmintic. The aim of this study is to know the efficacy of Mimba leaf (Azadirachta indica Less) ethanol extract was anthelmintic againts Ascaris suum in vitro. This study used Ascaris suum instead of Ascaris lumbricoides, because the high similarity between them and it was much easier to get Ascaris suum. This study was laboratory experimental with the post only controlled group design. The sample was divided into five groups: negative control group (NaCl 0.9 %), positive control group (pyrantel pamoat solution 1%), and experimental groups with several concentrations of mimba leaf extract (20%, 30%, and 40%). The worms were observed in 12 hours starts in 1st hours, 2nd, 3rd ,4th, 5th, 6th, 7th, 8th, 9th,10th,11th, until 24th hours. Data were analyzed by Probit analysis using IBM SPSS 22.0. The result showed that mimba leaf ethanol extract has Lethal Concentration 100 (LC100) 34,7%. From the result it can be concluded that Lethal Time 100 (LT100) of 40% extract mimba leaf was in 15 hours 9 minutes. This study shows that ethanol extract of mimba leaf (Azadirachta indica Less) has the effect of anthelmintic againts Ascaris suum in vitro.
