Abstract

Di Negara berkembang diare merupakan masalah utama penyebab kematian, termasuk Indonesia. Bakteri dari genus Shigella diketahui sebagai salah satu agen penyebab diare. Setiap tahun semakin banyak pelaporan kasus kurangnya efektifitas antibiotik, oleh karena itu diperlukan metode baru dalam penanganan diare salah satunya adalah dengan menggunakan vaksin. Vaksin yang banyak diteliti akhir-akhir ini adalah vaksin yang bertujuan mencegah tingkat keparahan penyakit atau vaksin yang bertujuan mencegah perlekatan pathogen. Penelitian ini bertujuan untuk mengetahui apakah ada proses adhesi antara protein adhesin pili 49,8 kDa Shigella sonnei dengan Enterocyte Mencit Balb-c. Metode yang digunakan adalah dengan metode pewarnaan Imunositokimia pada slide dengan pengenceran protein 1/500, 1/2000, 1/8000, dan kontrol negatif (tanpa antibodi) dan pengenceran antibodi 1/1600. Kemudian dilakukan penghitungan dari hasil Imunositokimia pada mikroskop dengan metode penghitungan 10 lapang pandang dengan pengulangan sebanyak 4x. Nilai-nilai rerata dari penghitungan tersebut kemudian dianalisis dengan uji statistik Anova, korelasi, dan regresi. Hasil uji Anova menunjukan adanya perbedaan bermakna tiap-tiap konsentrasi pengenceran dengan tingkat kepercayaan sebesar 95% (α<0,05). Uji korelasi dan regresi menunjukkan bahwa dengan meningkatnya konsentrasi pengenceran maka jumlah protein yang menempel pada enterosit semakin meningkat. Maka dapat disimpulkan bahwa ada proses adhesi antara protein adhesin pili Shigella sonnei 49,8 kDa dengan enterosit.

English Abstract

In developing countries including Indonesia, diarrhea is a major cause of death. Bacteria of the genus Shigella is known as one of the causative agents of diarrhea. Every year, there has been more and more reporting cases about the lack of effectiveness of antibiotics, therefore, new methods of treatment of diarrhea are required; one of which is the use of vaccines. The vaccine that is recently widely investigated is a vaccine intended to prevent the severity of the disease or prevent the attachment of pathogens. This study aims to determine if there is a process of adhesion between adhesion pili 49.8 kDa protein of Shigella sonnei and Balb-c mice enterocyte. The method used is the immunocytochemistry staining on slides with protein dilution 1/500, 1/2000, 1/8000, and a negative control (no antibody) and antibody dilution of 1/1600. Then, calculation was done from the results of microscopy immunocytochemistry by the method of calculating 10 fields of view with repetition as much as 4x. Mean values of these calculations were then analyzed with ANOVA statistical test, correlation test, and regression test. Results of ANOVA test showed significant differences in the concentration of each dilution with a confidence level of 95% (α<0,05). Correlation and regression tests showed that with increasing concentrations of dilution, the number of proteins that attach to the enterocytes increased. Therefore, it can be concluded that there is a process of adhesion between Shigella sonnei pili adhesin protein of 49.8 kDa and enterocytes.
