Abstract
Daun melati diketahui mengandung saponin, tannin dan flavonoid yang berperan dalam proses penyembuhan luka. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian ekstrak etanol daun melati dalam menurunkan jumlah makrofag luka bakar derajat II A. penelitian ini menggunakan design true experimental laboratory dengan metode Randomized Posttest Olny Controlled Group Design. Sampel yang digunakan dalam penelitian berjumlah 25 ekor tikus putih yang dibagi dalam 5 kelompok (n=5) yang terdiri dari 2 kelompok kontrol (Normal Saline 0.9%- SSD 1%) dan 3 kelompok perlakuan (EEDM 15%, 30% dan 45%). Perawatan dilakukan selama 14 hari dengan EEDM yang diberikan secara topical pada luka (luas 2x2 cm2). Pengambilan preparat dilakukan pada hari ke-15 dan dilakukan pengamatan histo menggunakan pewarnaan IHK di bawah mikroskop OLYMPUS seri CX 21, kamera digital Canon Ixus 105 dan software OlyVIA dengan perbesaran 400 kali dimana setiap sediaan diamati pada luas pandang 10 area. Didapatkan hasil rata-rata jumlah makrofag terendah pada kelompok perlakuan EEDM 30% (1.14±.13416) dan rata-rata jumlah makrofag tertinggi pada kelompok perlakuan EEDM 45% (2.3±.48391) dimana menunjukkan adanya pengaruh ekstrak daun melati terhadap jumlah makrofag pada penyembuhan luka bakar. Analisa data post hoc menunjukkan kelompok perlakuan EEDM 30% berpengaruh signifikan (p = 0.042) dibandingkan dengan kelompok kontrol negatif (Normal Saline 0.9%). Kesimpulan penelitian ini yaitu perawatan luka bakar derajat II A dengan ekstrak etanol daun sirih (Jasminum sambac Linn.) dengan konsentrasi 30% dapat menurunkan jumlah makrofag pada luka bakar derajat II A pada tikus putih (Rattus norvegicus) galur wistar.
English Abstract
Jasmine leaves are known to contain saponins , tannins and flavonoids play a role in the wound healing process. The purpose of this study was to determine the effect of ethanol extract of leaves of jasmine in reducing the number of macrophages in the second A degree burns wound. This study using true experimental design laboratory, methods Posttest Olny Controlled Group Design. The sample used in the study totaled 25 white rats were divided into 5 groups (n=5) consisting of 2 control groups (Normal Saline 0.9%-SSD 1%) and 3 treatment groups (EEDM 15%, 30% and 45%). Treatment was done for 14 days with EEDM given topically on wounds (width 2x2 cm2). Sample was taken on day 15th and were observed histo using IHK staining under the microscope OLYMPUS CX 21 series, the Canon Ixus 105 digital camera and software OlyVIA with magnification 400 where every sample inspected in 10 different area vast looks. The result showed the mean of macrophages in the treatment group EEDM 30% (1.14±.13416) and the mean of macrophages in the treatment group EEDM 45% (2.3±.48391) that show there is extracts ethanol of jasmine leaf has an effect on the number of macrophages in the healing of burns. Post-hoc test analysis of the data showed that treatment group EEDM 30% has significant effect (p = 0.042) compared with the negative control group (Normal Saline 0.9%). The conclusion of this study is that extract ethanol of Jasminum sambca leaf with concentration 30% can decrease the number of macrophages on second degree burn wound healing in Rattus norvegicus wistar strain.
