Abstract

Diabetes mellitus merupakan penyakit degeneratif dimana terjadi kondisi hiperglikemia kronis yang mampu mengakibatkan berbagai macam komplikasi yang dipicu oleh Advanced Glycation End Product (AGE). Selama ini terapi menggunakan aminoguanidine dan crosslinker lainnya membutuhkan terapi jangka panjang dan mahal sehingga pengembangan vaksinasi akan menjanjikan untuk terapi di masa depan. Penelitian ini menggunakan rancangan Randomized Posttest Only Controlled Group Design. Penelitian ini menggunakan 40 ekor mencit sebagai hewan coba, yang dibagi ke dalam 5 kelompok penelitian, kelompok kontrol negatif, kontrol positif, vaksinasi AGE-BSA, vaksinasi KLH, dan vaksinasi AGE-BSA-KLH. Hasil penelitian menunjukkan bahwa vaksinasi menggunakan AGE-BSA, KLH maupun AGE-BSA-KLH menghambat peningkatan kadar AGE yang signifikan (p=0.000; p<0.05) dengan korelasi yang sangat kuat (r=-0.925) dan pengaruh vaksinasi terhadap penghambatan peningkatan AGE sebesar 84%, peningkatan anti-AGE (p=0.000; p<0.05), penghambatan peningkatan MDA (p=0.000; p<0.05), penurunan berat liver (p=0.000; p<0.05) dan penghambatan peningkatan glukosa darah pasca mencit divaksinasi dan didiabeteskan (p=0.000; p<0.05). Penelitian ini menunjukkan bahwa vaksinasi menggunakan AGE-BSA, KLH dan AGE-BSA-KLH berpotensi untuk menurunkan faktor pemicu komplikasi pada diabetes mellitus melalui penghambatan pembentukan AGE.

English Abstract

Diabetes mellitus is one of the degenerative disease, with chronic hyperglycemia that will lead into increase of Advanced Glycation End Product (AGE). To date, aminoguanidine and other crosslinker agent to inhibit AGE need to be consumed forever. Therefore, vaccine development held promising potential in the future. This research used Randomized Posttest Only Controlled Group Design with 40 Balb/C mice as experimental animals, which is divided into 5 study groups, negative control, positive control, vaccination with AGE, vaccination with KLH (Keyhole Limpet Hemocyanin), and vaccination with AGE-KLH conjugates. The research results show that vaccination using AGE, KLH or AGE-KLH inhibit the increase of AGE significantly (p = 0.000, p <0.05) with a very strong correlation (r=-0925). Vaccination using AGE, KLH, or AGE-KLH conjugates will able increase anti-AGE level significantly (p = 0.000, p <0.05), inhibit the increase in MDA level(p = 0.000, p <0.05), decrease liver weight(p = 0.000, p <0.05) and inhibit the increase of blood glucose level (p = 0.000, p <0.05). Therefore,vaccination using AGE, KLH and AGE-KLH conjugates may be a potential approach to prevent diabetic complication through inhibition of AGE formation.
