Abstract
Streptococcus pyogenes merupakan salah satu bakteri yang dapat menyebabkan inflamasi lanjutan setelah terjadinya pulpitis pada gigi. Untuk mengatasi penyakit infeksi tersebut, banyak dikembangkan penggunaan antibakteri, termasuk antibakteri herbal. Salah satunya adalah daun cengkeh yang telah diketahui mengandung minyak atsiri dan flavonoid yang memiliki efek antibakteri. Penelitian ini bertujuan untuk membuktikan bahwa ekstrak daun cengkeh memiliki efek antibakteri terhadap Streptococcus pyogenes secara in vitro. Penelitian ini merupakan suatu penelitian eksperimental yang dilakukan terhadap Streptococcus pyogenes dengan metode dilusi tabung. Kelompok perlakuan yaitu kelompok bakteri yang diberi ekstrak daun cengkeh dengan konsentrasi 0.8%, 1%, 1.2%, 1.4%, dan 1.6%. Kelompok kontrol terdiri dari kontrol bakteri sebagai kontrol positif dan kontrol ekstrak daun cengkeh sebagai kontrol negatif. Hasil penelitian menunjukkan bahwa Kadar Hambat Minimal (KHM) diperoleh pada konsentrasi 1%, sedangkan Kadar Bunuh Minimal (KBM) diperoleh pada 1,6%. Analisa data menggunakan One-way ANOVA dengan α = 0,05 menunjukkan terdapat perbedaan efek antibakteri diantara tiap perlakuan konsentrasi ekstrak daun cengkeh terhadap jumlah koloni Streptococcus pyogenes. Uji korelasi Pearson menunjukkan semakin tinggi konsentrasi ekstrak daun cengkeh, maka semakin rendah pertumbuhan Streptococcus pyogenes secara in vitro. Kesimpulan pada penelitian ini yaitu ekstrak daun cengkeh mempunyai efek antibakteri terhadap Streptococcus pyogenes, dengan KBM 1,6%.
English Abstract
Streptococcus pyogenes is one of bacteria that cause exacerbation after the occurrence of pulpitis in a tooth. In order to eliminate the infection, antibacterial agents are developed, including those of herbal origins. One of the herb used as an antibacterial agent is clove leaf which has been known to contain essential oil, flavonoid and saponin. This research was conducted to prove the antibacterial effect of clove leaves extract on Streptococcus pyogenes using in vitro method. This research was an experimental study which was carried out to Streptococcus pyogenes with tube dilution method. The treated groups were bacteria which were given the clove leaves extract with range of concentration as follows 0.8%, 1%, 1.2%, 1.4%, and 1.6%. The control groups includes bacteria as positive control and clove leaves extract as negative control. The result indicated that minimal inhibitory concentration (MIC) was found at 1%, meanwhile minimal bactericidal concentration (MBC) was found at 1.6%. Results of One-way ANOVA with α = 0,05, showed significant difference among each concentration of clove leaves extract related to the growth of colonies of Streptococcus pyogenes. Pearson correlation test result showed that the higher concentration of clove leaves extract, the less number of colony growth. The conclusion of this study is that clove leaves extract has an antibacterial effect against Streptococcus pyogenes, with MBC is 1.6%.
