Abstract

Indonesia menempati peringkat ke-7 di dunia sebagai negara dengan jumlah penderita diabetes melitus terbanyak yang berjumlah 5 juta orang pada tahun 1998 dan diperkirakan tahun 2030 didapatkan 21,3 juta penderita diabetes melitus. Pada penyakit Diabetes Melitus tipe 1 (DM tipe 1), terjadi kerusakan sel islet langerhans yang mengakibatkan penurunan sekresi insulin yang ditandai dengan keadaan hiperglikemi kronis. Peran sel beta dalam perjalanan penyakit diabetes sangatlah penting. Saccharomyces cerevisiae merupakan salah satu ragi yang memiliki kandungan beta glucan yang tinggi. Beta glucan berperan dalam menstimulasi Hematopoietic Stem Cell (HSC) ke sirkulasi darah perifer. Mekanisme beta glucan dalam mempromosikan mobilisasi granulosit pada bone marrow menyebabkan regulasi ekspresi kemokin CXCR-4 dan SDF-1 turun sehingga memutuskan ikatan CD34 (HSC) dengan sel osteoblast lineage. Beta glucan juga mempengaruhi peningkatan granulosit dan mobilisasi granulosit serta progenitornya dengan menstimulasi produksi Granulocyte Colony Stimulating Factor (G-CSF) yang berperan dalam mobilisasi HSC ke sirkulasi perifer. Pemberian ragi Saccharomyces cerevisiae dengan dosis 100 mg/kgBB dan 200 mg/kgBB selama 6 minggu mampu meningkatkan jumlah sel beta pankreas (r = 0,926) dan menghambat gejala klasik DM tipe 1, yaitu penurunan berat badan secara signifikan (p < 0,05) dibanding dengan mencit yang diinduksi DM tipe 1 dengan menggunakan STZ. Pemberian ragi Saccharomyces cerevisiae dengan dosis 200 mg/kgBB mampu menurunkan kadar glukosa darah puasa secara signifikan dengan arah korelasi negatif yang kuat (r = -0,619), sedangkan pemberian dosis 50 mg/kgBB sudah mampu meningkatkan ekspresi sel CD34 secara signifikan (p < 0,05) pada sirkulasi perifer.

English Abstract

Indonesia ranks 7th in the world as the country with the highest number of people with diabetes mellitus, amounting to 5 million people in 1998 and in 2030 earned an estimated 21,3 million people with diabetes mellitus. On disease type 1 diabetes mellitus (Type 1 DM), Langerhans islet cell damage resulting in decreased insulin secretion characterized by a state of chronic hyperglycemia. The role of beta cells in the course of diabetes is very important. Saccharomyces cerevisiae is a yeast that has a high content of beta glucan. Beta glucan plays a role in stimulating the Hematopoietic Stem Cell (HSC) to peripheral blood circulation. The mechanism of beta glucan in promoting the mobilization of granulocytes in the bone marrow causes the regulation of chemokine expression of CXCR - 4 and SDF - 1 down so decided the bonding of CD34 (HSC) with osteoblast lineage cells. Beta glucan also affects the increase in granulocyte and granulocyte mobilization and the progenitor by stimulating the production of Granulocyte Colony Stimulating Factor (G - CSF) that play a role in HSC mobilization into peripheral circulation. Giving the yeast Saccharomyces cerevisiae with a dose of 100 mg/kg body weight and 200 mg/kg body weight for 6 weeks were able to increase the number of pancreatic beta cells (r = 0.926) and inhibits the classical symptoms of diabetes type 1, in example weight loss was significantly (p < 0.05) compared with mice induced type 1 diabetes using STZ. Giving the yeast Saccharomyces cerevisiae with a dose of 200 mg/ kg body weight can lower fasting blood glucose levels significantly with the direction of a strong negative correlation (r = -0.619), whereas the dose of 50 mg/kg body weight have been able to increase the expression of CD34 cells was significantly (p < 0 , 05) in the peripheral circulation.
