Abstract

Endosulfan adalah pestisida golongan cyclodiene. Pestisida jenis endosulfan paling banyak digunakan untuk mengontrol hama pada hasil panen seperti kopi, teh, kapas, dan hasil panen lainnya. Endosulfan masih banyak digunakan pada bidang pertanian dan kesehatan publik. Toksisitas endosulfan telah terbukti pada berbagai macam organ seperti otak, ginjal, liver, jantung, dan organ reproduksi. Zat ini dapat masuk kedalam tubuh melalui traktus intestinal, paru-paru, dan kulit. Pada tahun 2007 suatu penelitian menunjukkan bahwa ada hubungan antara paparan insektisida organoklorin pada ibu hamil selama periode organogenesis dengan perkembangan sistem syaraf. Endosulfan juga dapat mengganggu sistem imun seperti pembentukan sitokin pro inflamasi dan autoimun Penelitian ini bertujuan untuk mengetahui bahwa pemberian endosulfan berpengaruh terhadap kadar IL-17 serum tikus (Rattus norvegicus) strain wistar. Studi ini menggunakan metode randomized post test only controlled group design. Sampel dipilih secara acak kemudian dibagi menjadi 4 kelompok dengan 5 pengulangan yakni kelompok kontrol negatif, kelompok 1 dengan dosis endosulfan 0,07 mL, kelompok 2 dengan dosis endosulfan 0,33 mL, dan kelompok 3 dengan dosis endosulfan 1,6 mL. Pemberian endosulfan pada tikus sebanyak 7 kali pada usia kehamilan 6-20 hari. Kemudian setelah tikus melahirkan, anak tikus yang sudah berumur 4 minggu dikorbankan untuk diambil darahnya dari jantung. Hasil perhitungan IL-17 dengan metode ELISA menunjukkan bahwa pada kelompok tikus yang diberi paparan endosulfan menunjukan peningkatan kadar IL-17 secara bertahap setiap peningkatan dosis endosulfan (P1= 18,35 ± 7,61 ng/mL, P2= 26,67 ± 4,69 ng/mL dan P3= 30,72 ± 8,58 ng/mL). Peningkatan mencapai signifikansi (p<0,05). Dari hasil penelitian ini, terdapat hubungan antara peningkatan dosis endosulfan dengan kadar IL-17 (Uji Korelasi Pearson R=-0,509, p<0,05). Berdasarkan hasil penelitian ini, disimpulkan bahwa pemberian endosulfan mampu meningkatkan kadar IL-17 serum secara signifikan pada tikus Wistar dengan dosis terbesar 1,6 mL.

English Abstract

Endosulfan is a cyclodiene pesticide group. Endosulfan pesticide group is the most pesticide that widely used to control pests in crops such as coffe, tea, cotton, and other crops. Toxicity of endosulfan has been proven in many organs such as brain, kidney, liver, heart, and reproductive organs. These substance can poisoning the body through intestinal tract, lungs, and skin. In 2007, a study showed that there is relation between exposure of organochlorine pesticide in pregnant woman during the nerve development organogenesis period. Endosulfan can also interfere the immune system such as the formation of pro- inflammatory cytokines and autoimmunity. The purpose of this study was to prove that endosulfan could effect on serum levels of IL-17 rats (Rattus norvegicus) wistar strain. This experimental study used randomized post-test only group design. The subject were randomly selected and divided into 4 groups with 5 samples each group. the group were negative control group, group 1 at a dose of 0.07 mL endosulfan, endosulfan group 2 with 0.33 mL dose, and group 3 with 1.6 mL doses of endosulfan. Provision of endosulfan in rats as much as 7 times on days 6-20 of gestation. Then after the mice gave birth, rat pups 4 weeks old were sacrificed for blood drawn from the heart.The results of IL-17 serum calculated with ELISA method indicated that in the group of rats that were given endosulfan showed increased levels of IL-17 gradually increasing doses of endosulfan (P1= 18,35 ± 7,61 ng/mL, P2= 26,67 ± 4,69 ng/mL dan P3= 30,72 ± 8,58 ng/mL). Increased level of IL-17 serum reached significance difference (p<0,05). there was positive correlation between dose endosulfan an level IL-17 serum (Pearson correlation test R = 0,509, p<0,05). It is concluded that endosulfan can significantly increase levels of IL-17 serum of Wistar rats at a dose of 1.6 mL.
