Abstract
Streptococcus mutans merupakan bakteri rongga mulut yang menghasilkan zat asam yang dapat menimbulkan karies gigi. Pembersihan gigi secara mekanik menggunakan sikat dan pasta sering dilakukan untuk mencegah pembentukan plak bakteri penyebab karies gigi. Penggunaan daun asam sebagai bahan baku pasta gigi dapat menjadi alternatif pencegahan karies gigi. Daun asam (Tamarindus indica Linn.) mengandung zat antibakteri berupa flavonoid, tanin, saponin, dan alkaloid. Tujuan penelitian ini adalah untuk membuktikan bahwa ekstrak etanol daun asam memiliki efek antibakteri terhadap Streptococcus mutans secara in vitro. Penelitian ini merupakan eksperimental laboratorik dengan metode dilusi agar untuk mendapatkan nilai KHM dan dilusi tabung untuk mendapatkan nilai KBM. Konsentrasi ekstrak etanol daun asam yang digunakan adalah 2%, 4%, 6%, 8%, dan 10%. Hasil penelitian menunjukkan Kadar Hambat Minimum (KHM) diperoleh pada konsentrasi 4%, sedangkan Kadar Bunuh Minimum (KBM) pada konsentrasi 10%. Analisis data menggunakan oneway ANOVA menunjukkan adanya perbedaan yang signifikan pada perubahan konsentrasi ekstrak etanol daun asam terhadap jumlah koloni Streptococcus mutans (p<0,01). Uji korelasi Pearson menunjukkan adanya hubungan yang kuat dan berbanding terbalik antara konsentrasi ekstrak etanol daun asam dengan jumlah koloni Streptococcus mutans (-0,717). Uji regresi menunjukkan efektivitas ekstrak etanol daun asam terhadap Streptococcus mutans sebesar 51,4% (R square = 0,514). Berdasarkan penelitian ini, dapat disimpulkan bahwa ekstrak etanol daun asam mempunyai efek antibakteri terhadap Streptococcus mutans secara in vitro.
English Abstract
Streptococcus mutans is an oral bacterium that produces acid causing dental caries. Mechanical cleaning of teeth using brushes and paste is often done to prevent the formation of bacteria plaque causing dental caries. The use of tamarind leaves as a raw material of the toothpaste can be alternative prevention of dental caries. Tamarind (Tamarindus indica Linn.) leaves contain antibacterial substances such as flavonoids, tannins, saponins, and alkaloids. The purpose of this study is to prove that the ethanolic extract of tamarid leaves have antibacterial effects against Streptococcus mutans in vitro. This is a laboratory experimental study using agar dilution method to find MIC and tube dilution method to find MBC. The used concentration of ethanolic extract of tamarind leaves are 2%, 4%, 6%, 8%, and 10%. The result shows the Minimum Inhibitory Concentration (MIC) is concentration of 4%, while the Minimum Bactericidal Concentration (MBC) is concentration of 10%. Analyzing oneway ANOVA shows a significant difference in the change of concentration of ethanolic extract of tamarind leaves on the number of colonies of Streptococcus mutans (p <0.01). Pearson correlation test shows a strong and inversely relationship between the concentration of ethanolic extract of tamarind leaves and the number of colonies of Streptococcus mutans (-0,717). Regression test shows the effectiveness of ethanolic extract of tamarid leaves against Streptococcus mutans is 51.4% (R square = 0.514). Based on this study, it can be concluded that the ethanolic extract of tamarind leaves have antibacterial effects against Streptococcus mutans in vitro.
