Abstract
Oksidasi terhadap low density lipoprotein (LDL) menyebabkan terbentuknya reactive aldehyde, terutama malondialdehyde (MDA) dan menyebabkan LDL tidak dikenali oleh tubuh. Proses tersebut menyebabkan pembentukan sel busa dan peningkatan ketebalan aorta yang mengawali terjadinya aterosklerosis. Porphyromonas gingivalis mengekspresikan arginine-specific gingipain (Rgp) pada dinding sel bakteri tersebut yang mampu bereaksi silang dan berikatan dengan MDA-LDL. Tujuan dari penelitian ini adalah membuktikan bahwa vaksinasi dengan dinding sel bakteri P. gingivalis mampu menginduksi antibodi protektif terhadap aterosklerosis. Penelitian ini adalah eksperimen murni di laboratorium secara in vivo dengan metode Randomized Post Test Only Controlled Group Design. 25 tikus wistar jantan usia 8 minggu dibagi menjadi 5 kelompok yaitu kontrol positif, kontrol negatif, dinding sel-CFA/IFA 100 μl, CFA/IFA 100 μl, dinding sel 100 μl yang diberi diet atherogenik selama 30 hari dan diinjeksi vaksin 2 minggu sekali sebanyak 3 kali secara intraperitoneal. Tikus dibedah pada hari ke-31 dan diambil sampel darah serta aortanya. Uji spesifisitas antibodi yang dihasilkan dengan western blot dan aorta diambil untuk pengukuran ketebalan aorta dengan pengecatan hematoksilin eosin (HE). Pada penelitian ini vaksinasi dinding sel bakteri P. gingivalis mampu menginduksi pembentukan antibodi yang spesifik terhadap protein arginine specific gingipain (Rgp) pada dinding sel bakteri tersebut yang dapat bereaksi silang dan mengikat MDA-LDL dalam darah tikus. Hal tersebut dibuktikan dengan terjadi penurunan ketebalan dinding aorta tikus secara signifikan (p < 0,05). Berdasarkan hasil tersebut dapat disimpulkan bahwa vaksinasi dengan dinding sel bakteri P. gingivalis mampu menginduksi pembentukan antibodi spesifik yang bersifat protektif terhadap aterosklerosis secara in vivo.
English Abstract
The oxidation of low density lipoprotein (LDL) causes the formation of reactive aldehydes, especially malondialdehyde (MDA) and causes LDL can not be recognized by the body . The process causes the formation of foam cells and initiate the increasing of the thickness of the aortic that will initiate atherosclerosis formation. Porphyromonas gingivalis expresses arginine-specific gingipain (Rgp) on its cell wall that is capable to cross-react and bind to MDA-LDL. The purpose of this study is to prove that vaccination with bacterial cell wall of P. gingivalis is able to induce antibodies protective against atherosclerosis. This research is purely laboratory experiment in vivo with a method Randomized Controlled Group Post Test Only Design. 25 male Wistar rats aged 8 weeks are divided into 5 groups: positive control, negative control, wall cell-CFA/IFA 100 mL, CFA/IFA 100 mL, 100 mL wall cell fed atherogenic diet for 30 days and injected vaccine every 2 weeks intraperitoneally 3 times each. Rats are dissected on day 31, blood and aortic tissue are taken for samples. Western blot test is used to determine the specificity of the antibodies and hematoxylin eosin staining (HE) is used to know the thickness of the aorta. In this study the bacterial cell wall vaccination P. gingivalis is able to induce the formation of antibodies specific to the protein arginine-specific gingipain (Rgp) on the bacterial cell wall that can cross-react and bind to MDA-LDL in the blood of mice. This is proven by a decreasing of rat aortic wall thickness significantly (p < 0.05). Based on these results it can be concluded that vaccination with bacterial cell wall of P. gingivalis is able to induce the formation of specific antibodies that are protective against atherosclerosis in vivo.
