Abstract

Fibrosis hepar merupakan salah satu masalah kesehatan utama di dunia yang menyebabkan sekitar 1,5 juta kematian per tahun. Kematian ini sebagian besar disebabkan oleh stadium akhir dari fibrosis seperti sirosis hati dan kanker hepar (Naveau et al., 2009).Pada saat telah mencapai tahap sirosis, transplantasi hepar merupakan terapi yang paling efektif untuk penderita.Akan tetapi sangat sedikit sekali organ donor yang sesuai dengan penderita dan seringkali terjadi komplikasi. Dilain sisi kulit kacang tanah (Arachis hypogaea L.) kurang dimanfaatkan dan dianggap sebagai. Kandungan terbesar dari flavonoid itu adalah luteolin. Luteolin berperan sebagai antiinflamasi, antioksidan, dan antikanker. Pada hubungannya dengan fibrosis hepar, luteolin mampu menghambat aktivasi NFκB pada tahapan produksi TGF-β1 pada level sitosolik sehingga menghambat aktivasi HSCs yang menyebabkan penumpukan jaringan kolagen dan penurunan regenerasi sel hepar. Tujuan penelitian ini membuktikan pengaruh ekstrak kulit kacang terhadap penurunan kadar TGF-β1 hepar pada jaringan hepar mencit model fibrosis hepar. Penelitian ini merupakan penelitian murni dengan metode Randomized Posttest Only Controlled Group Design. Mencit BALB/c jantan sebagai hewan coba dibagi menjadi 5 kelompok, yaitu : kontrol positif, kontrol negatif, P1 (15 ml/KgBB), P2 (30 ml/KgBB), P3 (60 ml/KgBB) kemudian kontrol positif dan P1, P2, P3 diberi Induksi CCL4 selama 6 minggu hari dan diberi ekstrak mulai hari minggu ke-2 sampai minggu ke-6. Hasil penelitian menunjukkan bahwa ada penurunan kadar TGF-β1 pada jaringan hepar mencit model fibrosis hepar. Sehingga dapat disimpulkan bahwa ekstrak kulit kacang tanah mampu menurunkan kadar TGF-β1 pada jaringan hepar mencit Balb/c model fibrosis hepar.

English Abstract

Liver fibrosis is one of the major health problems in the world which causes about 1.5 million deaths per year. Death is largely due to the late stage of fibrosis such as liver cirrhosis and liver cancer ( Naveau et al., 2009). At the moment had reached the stage of cirrhosis, liver transplantation is the most effective therapy for patients. But very few organ donors accordance with the patient and complications occur frequently. On the other side of the shell peanuts ( Arachis hypogaea L. ) is still considered underutilized. The content of flavonoids was greatest luteolin in it. Luteolin acts as an anti-inflammatory, antioxidant, and anticancer. In conjunction with hepatic fibrosis, luteolin is able to inhibit the activation of NFκB in TGF - β1 production phases in cytosolic levels thereby inhibiting activation of HSCs that causes a buildup of collagen tissue and liver cell regeneration decrease. The purpose of this study demonstrates the effect of peanut hull extract reduced levels of TGF- β1 in liver tissue in mice model of hepatic fibrosis. This study is purely the method Randomized Controlled Group Posttest Only Design. BALB / c mice as experimental animals males were divided into 5 groups , namely : positive control , negative control , P1 ( 15 ml / kg ) , P2 ( 30 ml / kg ) , P3 ( 60 ml / kg ) and then the positive control and P1 , P2 , P3 given CCl4 for 6 weeks induction days and were given extracts from today week 2 to week 6. The results showed that there was decrease in levels of TGF - β1 in the liver tissues of mice model of hepatic fibrosis. It can be concluded that the peanut hull extract could reduce levels of TGF-β1 in liver tissue of Balb/cmice model of liver fibrosis.
