Abstract
Infeksi Staphylococcus aureus (S. aureus) saat ini menjadi masalah kesehatan yang serius. Bahkan beberapa galur tertentu dari bakteri ini mampu membentuk biofilm pada alat-alat medis seperti kateter urin, sendi prostetik, prosthetic heart valve, dan pacemaker. Dengan adanya biofilm, bakteri pembentuk biofilm bersifat sangat resisten terhadap bahaya dari lingkungan dan terapi antibiotika. Oleh karena itu, perlu dilakukan penelitian untuk mencari senyawa yang dapat menghambat pembentukan biofilm. Penelitian ini dilakukan untuk mengetahui pengaruh ekstrak mahkota bunga mawar dalam menghambat pembentukan biofilm S. aureus. Pemberian ekstrak mahkota bunga mawar yang mengandung tannin, terpene, dan flavonoid diharapkan dapat menghambat pembentukan biofilm pada S. aureus. Desain penelitian ini adalah true experiment post-test only group design. Sampel diperoleh dari swab tenggorok di Laboratorium Mikrobiologi FKUB. Konsentrasi ekstrak mahkota bunga mawar yang digunakan adalah 0% (kontrol), 15%, 20%, 25%, 30%, dan 35% kemudian dilakukan pengulangan 4 kali. Pembentukan biofilm diamati dengan metode tabung kemudian diukur mean gray value nya dengan program Adobe Photoshop CS3. Analisis data yang digunakan adalah Uji One Way ANOVA dan Uji Korelasi Pearson. Hasil statistik Uji One Way ANOVA menunjukkan adanya perbedaan yang signifikan pada perubahan konsentrasi ekstrak mahkota bunga mawar terhadap mean gray value (p < 0,05). Uji Korelasi Pearson menunjukkan adanya hubungan yang sangat kuat antara konsentrasi ekstrak dengan mean gray value (r = 0,885; p < 0,05). Dari hasil penelitian ini disimpulkan bahwa ekstrak mahkota bunga mawar dapat menghambat pembentukan biofilm dengan Minimum Biofilm Inhibitory Concentration (MBIC) sebesar 35%.
English Abstract
Nowadays, Staphylococcus aureus (S. aureus) infection has become a serious health problem. The more important thing is that some of these strains can form biofilm in medical devices such as urinary catheter, prosthetic joint, prosthetic heart valve, and pacemaker. With the presence of biofilm, biofilm forming bacteriae have resistant characteristics towards the hostile environment and antibiotics therapy. Therefore, it is necessary to find a compound that can inhibit biofilm formation. This study was conducted to prove the effect of rose petals extract as S. aureus biofilm formation inhibitor. The rose petals contain tannin, terpene, and flavonoid, and were expected to inhibit the growth of biofilm produced by S. aureus. This research was using post-test only group design. Isolates were taken from throat swab at Laboratorium of Microbiology Department, Faculty of Medicine, Brawijaya University. The concentrations of rose petals extract were 0% (control), 15%, 20%, 25%, 30%, and 35% then the experiment was repeated 4 times. The biofilm formation was observed with tube-test method, mean gray value was measured using Adobe Photoshop CS3 program. This study used One Way ANOVA test and Pearson test for statistical analytic tests. The result of One Way ANOVA showed that there was a significant difference in various extract’s concentrations given to S. aureus (p < 0,05). The result of Pearson test showed that there was a significant association between concentration of the extract and mean gray value (r = 0,885; p < 0,05). The conclusion is that the rose petals extract could inhibit S. aureus biofilm formation in vitro with the Minimum Biofilm Inhibitory Concentration (MBIC) at 35%.
