Abstract

Aterosklerosis merupakan penyebab berbagai penyakit kardiovaskuler yang telah dianggap sebagai masalah kesehatan utama di seluruh dunia. Banyak faktor yang berkontribusi terhadap perkembangan proses aterosklerosis. Salah satu faktor yang berperan penting dalam proses aterosklerosis adalah respon inflamasi. Respon inflamasi akan menyebabkan banyak mediator inflamasi yang dilepaskan, salah satunya adalah TNF-α. Salah satu upaya pencegahan aterosklerosis yang sedang banyak diteliti adalah dengan antosianin sebagai antioksidan. Antosianin banyak terdapat pada tumbuh-tumbuhan salah satunya adalah ubi jalar ungu yang diketahui mempunyai efek sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian antosianin dari ubi jalar (Ipomoea batatas L.) varietas ungu terhadap kadar TNF-α serum tikus wistar (Rattus norvegicus) yang diberi diet tinggi lemak. Studi eksperimental ini menggunakan desain penelitian post test only control group selama 94 hari. Sebanyak 25 ekor tikus dibagi secara acak menjadi 5 kelompok yang masing-masing terdiri atas 5 ekor tikus, yaitu kelompok kontrol negatif (diet normal), kontrol positif (diet tinggi lemak), perlakuan Fa (diet tinggi lemak + antosianin 5 mg/hari), perlakuan Fb (diet tinggi lemak + antosianin 10 mg/hari), dan perlakuan Fc (diet tinggi lemak + antosianin 20 mg/hari). Dari hasil uji statistik One Way ANOVA diketahui bahwa tidak terdapat perbedaan kadar TNF-α serum yang signifikan antar kelompok perlakuan (p=0,691). Dengan demikian, dapat disimpulkan bahwa dosis antosianin 5 mg/hari, 10 mg/hari, dan 20 mg/hari tidak dapat secara signifikan menghambat peningkatan kadar TNF-α serum tikus yang diberi diet tinggi lemak

English Abstract

Atherosclerosis is a cause of cardiovascular diseases which have been considered as a major health problem worldwide. Many factors have contributed to development of atherosclerosis. One of the important factor is the inflammatory response. The inflammatory response causes many inflammatory mediators are released, one of which is TNF-α. One of prevention of atherosclerosis which being widely studied is the anthocyanin as antioxidant. Anthocyanin found in many plants, one of which is the purple sweet potato that known widely for its antioxidant effect. This study aims to determine the effect of anthocyanin from purple sweet potato (Ipomoea batatas L.) on serum levels of TNF-α Wistar rats (Rattus norvegicus) fed with high fat diet. This experimental study is used post test only control group research design for 94 days. A total of 25 rats were randomly divided into 5 groups, each consisting of 5 rats, the negative control group (normal diet), positive control group (high fat diet), the treatment Fa (high fat diet + anthocyanin 5 mg / day) , treatment Fb (high fat diet + anthocyanin 10 mg / day), and treatment Fc (high fat diet + anthocyanin 20 mg / day). One way ANOVA test statistically indicated that there is no significant difference in serum levels of TNF-α between treatment groups (p = 0.691). Thus, it can be concluded that anthocyanin dose of 5 mg/day, 10 mg/day, and 20 mg/day did not significantly inhibit the increase of serum levels of TNF-α mice fed with high fat diet.
