Abstract
Oksidasiterhadap low density lipoprotein LDL menyebabkan terbentuknya reactive aldehyde, terutama malondialdehyde (MDA) dan menyebabkan LDL tidak dikenali oleh tubuh. Proses tersebut menyebabkan pembentukan sel busa dan peningkatan ketebalan aorta yang mengawali terjadinya aterosklerosis Porphyromonasgingivalis mengekspresikan arginine-specific gingipain (Rgp) pada dinding sel bakterit ersebut yang mampu bereaksi silang dan berikatan dengan MDA-LDL. Tujuan dari penelitian ini adalah mengetahui pengaruh pemberian vaksin aterosklerosis menggunakan dinding sel bakteri Porphyromonasgingivalis terhadap kadar MDA-LDL pada serum darah tikus wistar yang di induksi diet aterogenik. Penelitian ini adalah eksperimen murni di laboratorium secara in viv o dengan metode Randomized Post Test Only Controlled Group Design . 25 tikus wistar jantan usia 8 minggu di bagi menjadi 5 kelompok yaitu kontrolpositif, kontrolnegatif, dindingsel-CFA/IFA 100μl, CFA/IFA 100μl, dinding sel 100 μl yang diberi diet atherogenik selama30 hari dan di injeksi vaksin 2 minggu sekali sebanyak 3 kali secara intraperitoneal. Tikus di bedah pada hari ke-31 dan d ambil sampel darah serta aortanya.Uji spesifisitasanti bodi yang di hasilkan dengan western blot dan pengukuran kadar MDA-LDL dengan enzyme linked immunosorbent assay (ELISA).). Pada penelitian ini vaksinasi dinding sel bakteri P. gingivalis mampu menginduksi pembentukan anti bodi yang spesifik terhadap protein arginine specific gingipain (Rgp) pada dinding sel bakteri tersebut karena dapat bereaksi silang dan mengikat MDA-LDL dalam darah tikus. Hal tersebut dibuktikan dengan hasil uji anova yang menunjukkan penurunan kadar MDA-LDL serum tikus secara signifikan (p < 0,05). Berdasarkan hasil tersebut dapat disimpulkan bahwa pemberian vaksinasi menggunakan dinding sel bakteri P. Gingivalis mampu menurunkan kadar MDA-LDL serum tikus yang diinduksi aterosklerosis.
English Abstract
Atherosclerosis is a chronic inflammatory disease caused by lipid deposition in arterial wall. Oxidation of low-density lipoprotein (LDL) can cause the formation of reactive aldehydes, mainly malondealdehyde. This process will make the LDL can not be recognized by our body. It causes the foam cell formation and thickening of arterial wall which are the start of atherosclerosis process. Porphyromonas gingivalis expressing arginine-specific gingipain (RgP) induce anti–RgP antibody response which cross reacts with RgP in malondealdehyde modified lipoprotein (MDA-LDL). Anti-RgP antibody in turn will induce atheroprotective effect. The objective of this research was to know the effect of vaccination using Porphyromonas gingivalis against malondealdehyde modified low density lipoprotein (MDA-LDL) level in the sera of atherosclerosis rat model. This research was an experimental laboratory research with posttest only controlled group design. 25 male rats (8 weeks) were divided into 5 groups, negative control, positive control, first group which was vaccinated by Porphyromonas gingivalis cell wall and CFA-IFA, second group which was vaccinated by Porphyromonas gingivalis cell wall only, and the last group which were vaccinated by CFA-IFA only. We gave all groups except negative control the atherogenic diet for 30 days. Vaccine were injected intraperitoneallythree times at two-week interval. After 30 days, the rats were dissected to draw the blood for sample. The specificity of antibody was done by western-blot and enzyme linked immunosorbent assay (ELISA) was done for quantifying of MDA-LDL level. Results showed that administration of Porphyromonas gingivalis cell wall could induce antibody formation against arginine-specific gingipain (Rgp). Anova test showed the significant decrease of MDA-LDL level in the sera of atherosclerosis rats (p < 0,05). It means that the antibody could bind with MDA-LDL in rats blood. Based on the result, it can be concluded that vaccination using Porphyromonas gingivalis cell wall could decrease MDA-LDL level in the sera of atherosclerosis
