Abstract
Methicillin Resistant Staphylococcus aureus (MRSA) merupakan galur spesifik dari bakteri Staphylococcus aureus yang resisten terhadap antimikroba semua turunan penicillin dan methicillin serta antimikroba spektrum luas beta-lactamase resisten penicillin. Infeksi MRSA terbukti lebih berbahaya, dibandingkan kasus Staphylococcus biasa karena resisten terhadap pengobatan. Selain itu, bakteri MRSA merupakan penyebab penting dari infeksi nosokomial atau didapat dari rumah sakit. Oleh karena itu, perlu dikembangkan alternatif pengobatan sebagai solusi mengatasi masalah resistensi antibiotik. Salah satu tanaman yang telah banyak dimanfaatkan secara tradisional namun belum banyak diteliti aktivitas antibakterinya adalah Katuk (Sauropus androgynus). Penelitian ini bertujuan untuk membuktikan efek antibakteri ekstrak etanol daun katuk terhadap MRSA secara in vitro. Metode yang digunakan adalah uji dilusi tabung yang terdiri atas tahap penentuan Kadar Hambat Minimum (KHM) dan Kadar Bunuh Minimum (KBM). Konsentrasi ekstrak etanol daun katuk yang digunakan adalah 4% v/v, 6% v/v, 8% v/v, 10% v/v, 12% v/v, 14% v/v, dan 16% v/v, sedangkan konsentrasi MRSA adalah 106 CFU/ml. Hasil uji statistik menunjukkan bahwa ekstrak etanol daun katuk, secara signifikan dapat menghambat pertumbuhan MRSA (ANOVA, p = 0.000) dan terdapat hubungan antara peningkatan konsentrasi ekstrak etanol daun katuk dengan penurunan jumlah pertumbuhan koloni MRSA (R =-0.837, p = 0.000). KHM ekstrak etanol daun katuk terhadap MRSA adalah 14% v/v dan KBM-nya adalah 16% v/v. Kandungan ekstrak etanol daun katuk yang diperkirakan berperan sebagai antibakteri adalah saponin, flavonoid, dan tannin. Dengan demikian, dapat disimpulkan bahwa ekstrak etanol daun katuk memiliki efek antibakteri terhadap MRSA secara in vitro.
English Abstract
Methicillin Resistant Staphylococcus aureus (MRSA) is a specific strain of the Staphylococcus aureus that is resistant to antimicrobials such as derivates of penicillin, methicillin and antimicrobial broad spectrum beta-lactamase-resistant penicillin. MRSA infection shown to be more dangerous than ordinary Staphylococcus case as resistant to treatment. In addition, MRSA is an important cause of nosocomial infections or hospital acquired. Therefore, its necessary to develop alternative treatment as a solution to overcome the problem of antibiotic resistance. One plant that have been used traditionally but has not been widely studied its antibacterial activity is katuk (Sauropus androgynus). This experiment aims to verify the antibacterial effects of ethanolic extract of Katuk’s leaves against MRSA in vitro. The antibacterial test used in this experiment is the tube dilution test to determine the Minimum Inhibitory concentration (MIC) and Minimum Bactericide Concentration (MBC). The extract concentration used were 4%, 6%, 8%, 10%, 12%, 14%, and 16%, while the concentration of MRSA was 106 CFU/ ml. Statistical analysis showed that the ethanolic extract of Katuk’s leaves could significantly inhibit the growth of MRSA (ANOVA, p = 0.000) and there was a relationship between the increase ethanolic extract of Katuk’s leaves concentration with the decrease of MRSA colony numbers (R = -0837). The research also showed that the MIC was 14% v/v and MBC was 16% v/v. The main contents of ethanolic extract of Katuk’s leaves that is believed to be antibacterial agent are saponin, flavonoid, and tannin. It can be concluded that ethanolic extract of Katuk’s leaves has an antibacterial effect against MRSA in vitro
